摘  要:目的研究不同输出功率(w)和照射时间(t)设置下Nd:YAG激光照射对体外培养的人牙周膜成纤维细胞存活率的影响,并分析其相关性。方法将原代培养的人牙周膜成纤维细胞以3×103个/ml接种于96孔板,按不同输出功率和照射时间对细胞进行分组、激光照射,台盼兰染色和细胞存活率计算,采用spss15.0软件包,对实验组和对照组的细胞存活率进行成组设计资料的t检验(Independent Samples Ttest),对输出功率、照射时间和细胞成活率分别进行Pearson线性相关分析,对输出功率、照射时间和细胞成活率相关的显著性进行多元线性回归分析。结果 1w、10s组的细胞存活率与对照组无显著性差异(P>0.05),余实验组与对照组的差异均有统计学意义(P<0.05),且当激光的总能量>40J时,细胞存活率显著下降(P<0.001);照射时间为10s、20s和30s时,细胞存活率与输出功率呈负相关,20s时最为显著(r=-0.99);细胞存活率与照射时间负相关较输出功率显著(rt=-0.996,rW=-0.897)。结论 Nd:YAG激光照射可影响牙周膜成纤维细胞的存活,其中照射时间比输出功率对细胞存活率的影响更大。Objectives To evaluate the effect of the pulsed Nd:YAG laser irradiation on the survival rate of human periodontal ligament fibroblast at different power and irradiation time settings in vitro, and to analyze relevance among them. Methods The cells were cultured and inoculated into the 96-well tissue culture plates by 3×10^3per milliliter.Cells were divided into groups according to the irradiation time (10s,20s,30s, 40s) and the output power (1.0W,1.5W,2.0W).After the irradiation , cells were Stained with Trypan blue and calculated survival rates. Experimental groups and control group's dates were obtained and analyzed by Students't test and Linear Regression with spss 15.0 software package. Results No significant survival rates were found in between 1.0 w,10 s setting and control group(P〉0.05). The cell survival rate decreased significantly when laser energy were beyond 40J(P〈0.001). When exposure for 10s, 20s and 30s ,the cell survival rate and the output power was a negative correlation, the most significant was 20s (r=-0.99).Besides, Cell survival rates and exposure time were more significant negative correlation than the output power(rt=- 0.996, rW =-0.897). Conclusion Nd: YAG laser irradiation may affect the survival of periodontal ligament fibroblasts and speculates exposure time have greater impact on cell survival rate than output power.
