摘  要:目的研究瘦素(Leptin)对大鼠关节软骨的作用。方法经对SD大鼠膝关节注射瘦素后,取胫骨平台关节软骨,分别用RT-PCR法及免疫组织化学法检测胫骨平台中瘦素(Leptin)及转移生长因子β(TGF-β)的变化。结果瘦素无论在RNA水平或是蛋白水平都可以刺激正常关节软骨表达瘦素并可介导TGF-β的合成。结论该研究表明大鼠膝关节软骨可表达瘦素受体,瘦素在外周对关节软骨具有保护作用,并在骨关节炎的病理生理过程中起到重要的作用。【Objective】To determine whether rat articular chondrocytes express leptin receptor, and to determine whether leptin has a key role on rat articular chondrocytes. 【Methods】Leptin (100μg) was injected into the keen joints of rats. Tibial plateaus were collected and processed for transcriptase polymerase chain reaction and immuno- histochemical analysis. 【Results】Animal studies showed that leptin strongly stimulated anabolic functions of chondrocytes and induced the synthesis of TGF-β in rat cartilage at both the messenger RNA and the protein levels. 【Conclusion】Rat articular chondrocytes express leptin receptor. These findings suggest a new peripheral function of leptin as a key regulator of chondrocyte metabolism, and indicate that leptin may play an important role in the pathophysiology of OA.
