摘  要:目的探讨高脂膳食对学习记忆及突触可塑性的影响,为合理膳食预防脑老化提供科学依据。方法雄性ICR小鼠分为4组,即高脂膳食组、脑老化组、模型组、普通对照组,每组10只。以每天D-半乳糖100mg/kg皮下注射法制备脑老化模型,实验干预期共9w。以Morris水迷宫测试小鼠空间记忆和学习能力,流式细胞术测定皮层海马神经元突触体数量,荧光偏振法测定突触体膜流动性。结果①水迷宫实验中,脑老化组小鼠的逃避潜伏期(EL)明显大于普通对照组小鼠(P<0.05),模型组小鼠的EL与脑老化组无显著差异(P>0.05),高脂膳食组与普通对照组小鼠的EL无显著差异(P>0.05)。②突触体数量:脑老化组和模型组小鼠低于非脑老化组(高脂膳食组和普通对照组,P<0.05),模型组与脑老化组之间无显著差异(P>0.05),但高脂膳食组显著低于普通对照组(P<0.05)。③突触体膜流动性:脑老化组和模型组小鼠显著低于普通对照组(P<0.05)。脑老化组和模型组之间无显著差异(P>0.05)。高脂膳食组显著低于普通对照组(P<0.05)。结论高脂膳食可使小鼠皮层海马突触体数量减少,突触体膜流动性降低,但对小鼠学习记忆能力未见明显影响,对D-半乳糖诱导的脑老化也无明显协同效应。Objective To examine the effects of high fat diet on learning as well as on synaptic plasticity and afford scientific evidence to rational diet and prevent brain aging. Methods Forty male ICR mice were randomly divided into the high fat diet only, the D-galactose-induced brain aging, brain aging plus high fat diet, and normal control groups. Mice were given 100 mg ·kg^-1 · d^-1 subcutaneous injection of D-galactose to prepare brain aging model for 9 w. Morris water maze (MWM) test was employed to determine their spatial learn- ing and memory abilities. Flow cytometry (FCM) was used to analyze the amount of hippocampal synaptosomes, Membrane fluidity of synaptosomes was measured by fluorescence polarization technique. Results ( 1 ) In Morris water maze test, brain aging mice showed a signifieanly longer escape latency (EL) than that of the normal control mice ( P 〈 0. 05 ). There was no statistical difference in EL between brain aging mice plus high fat diet and brain aging mice groups ( P 〉 0. 05 ), and between the control and high fat diet groups ( P 〉 0. 05 ). ( 2 ) Number of synaptosomes: The number of synaptosomes in brain aging mice and brain aging mice plus high fat diet groups were less than those in nonbrain aging mice (the high fat diet and the control mice groups) (P 〈 0.05 ). There were no statistical difference in the number of synapto- somes between brain aging mice plus high fat diet and brain aging mice groups(P 〉0. 05). The high fat diet group was less than that of the normal control group ( P 〈 0. 05 ). ( 3 ) Membrane fluidity of synaptosomes : The fluidity of membrane in brain aging group and brain aging mice plus high fat diet group were less than that in the control group ( P 〈 0. 05 ). There were no statistical difference between brain aging mice plus high fat diet groups and brain aging mice group ( P 〉 0. 05 ).
