摘  要:目的探讨促血管生成素2(Ang-2)及血管内皮生长因子(VEGF)在肠腺瘤组织中的表达,研究其在肠腺瘤进展至腺癌中的作用及其临床意义。方法采用免疫组织化学方法检测Ang-2、VEGF和ki67蛋白在30例肠腺瘤中的表达水平,计数微血管密度(MVD)。结果 30例肠腺瘤,其中Ang-2阳性表达率为40%,VEGF为30%,Ang-2和VEGF的阳性表达率显著高于正常对照组(P=0.041,0.036),小于大肠癌组(P=0.012,0.020);肠腺瘤中MVD和ki67的表达明显高于正常对照组(P=0.018,0.034),低于大肠癌组(P=0.023,0.042)。肠腺瘤中Morson分级高和多发性息肉中Ang-2的表达高于分级低和单发性息肉;腺瘤直径大、Morson分级高和多发性息肉的VEGF表达高于直径小、分级低和单发性息肉;腺瘤直径大、Morson分级高的MVD的表达高于直径小和分级低者。结论 Ang-2、VEGF及其相互作用在肠腺瘤进展至腺癌中可促进肿瘤血管生成,可能在大肠腺瘤癌变中起到重要作用。ObjectiveWe explored the role of expressions of angiopoietin-2 (Ang-2) and vascular endo thelial growth factor (VEGF) in tumor angiogenesis in human colon adenamas and analyzed their relationship with clinicle pathological characteristics.MethodsImmunohistochemistry was used to examine the expression of Ang-2, microvessel density (MVD) and VEGF in 30 cases with colon adenama and 58 cases with colon cancer.ResultsThe positive percentage of Ang-2(40%) and VEGF(30%) in colon adenamas was higher than that in control(P=0.041, 0.036), however, lower than that in colon cancer (P=0.012, 0.020). The expression of MVD and ki67 was higher than that in control(P=0.018, 0.034), but lower than that in colon cancer (P=0.023, 0.042). The expression of Ang-2 in high Morson stages and multiple polypers was higher than that in low Morson stages and single polypers. The expression of VEGF in high Morson stages, large and multiple polypers adenomas was higher than that in low Morson stages, small and single polypers. The expression of MVD in high Morson stages and large polypers adenomas was higher than that in low Morson stages, small adenomas.ConclusionThe result of over-expression of Ang-2, VEGF and their interaction in human adenama may show higher tumor angiogenesis. Ang-2 and VEGF may play an important role in tumor progression and angiogenesis in colon adenama.
