异丙酚对局灶性癫痫大鼠发作期皮质脑电图棘波活动的影响

摘　　要：目的　观察异丙酚对青霉素 G诱发局灶性癫痫大鼠发作期皮质脑电图 (ECo G)棘波活动的影响。方法　 SD大鼠 40只 ,青霉素 G制成局灶性癫痫大鼠模型 (棘波频率在 30～ 5 0个棘波 /min) ,随机分为 5组 ,每组 8只。 组为对照组 ,其余大鼠分别持续静脉注射 30 ( 组 )、6 0 ( 组 )、90( 组 )、12 0 ( 组 ) mg· kg- 1 · h- 1 异丙酚 30分。测定异丙酚持续静脉注射 30分时的血药浓度 ,观察大鼠青霉素病灶区 ECo G发作期棘波频率 (F)的变化 ,探讨异丙酚血药浓度与青霉素 G诱发的局灶性癫痫大鼠病灶区 ECo G发作期棘波频率抑制效应〔IE=(F1 - Fn) / F1 ;n= 、 、 、 )〕的关系。结果　 ～ 组大鼠异丙酚持续静脉注射 30分时平均血药浓度分别为 (2 .9± 0 .3)、(5 .4± 0 .7)、(9.2± 1.2 )、(11.5± 0 .6 )μg· ml- 1 ;与对照组相比 , 、 组大鼠青霉素病灶区 ECo G发作间棘波频率无变化 , 组明显抑制 (P<0 .0 5 ) , 组显著抑制 (P<0 .0 1)。其相关系数 r=0 .97(P<0 .0 5 )。结论　异丙酚对青霉素 G诱发的局灶性癫痫大鼠病灶区 ECo G发作期棘波频率的抑制效应呈剂量依赖性。Objective To investigate the effect of propofol on interictal electrocorticographic(ECoG)spikes activity in rats with focal epilepsy.Methods Forty Sprague Dawley focal epileptic rats with interictal ECoG spikes(30 50spikes/min)induced by penicillin G,were randomly allocated into 5 groups,group I:control;in other groups,the rats were given 30(group Ⅱ),60(group Ⅲ) 90(group Ⅳ) and 120(group Ⅴ)mg·kg -1 ·h -1 of propofol for 30 min,respectively(n=8,in each group).After 30 min infusion of propofol,the changes of frequency of interictal spikes(F) were observed;the plasma concentration of propofol were measured,and the relationship between the plasma concentration of propofol and the inhibitory effect of interictal spikes(IE=[F 1-F n]/F 1;n=Ⅱ,Ⅲ,Ⅳ,Ⅴ)was observed.Results After 30min infusion of propofol,the average plasma concentration of propofol in group Ⅱ Ⅴ were (2.9±0.3),(5.4±0.7),(9.2±1.2),(11.5±0.6)μ g·ml -1 respectively.Compared with that in group Ⅰ,F in penicillin G focus had no changes in group Ⅱ and group Ⅲ,decreased in group Ⅳ(P<0.05),decreased significantly in group Ⅴ(P<0 01).The inhibitory effect of F in rats with focal epilepsy induced by penicillin G postively related to the plasma concentration of propofol(r=0 97,P<0 05) Conclusion Propofol may inhibit the frequency of interictal spikes of focal epilepsy in dose related way
