摘 要:将携带有人粒细胞-巨噬细胞集落刺激因子(GM-CSF)基因的重组腺病毒载体转染BALB/C小鼠肝癌细胞株(H22),体外经60Cγ射线灭活后制成瘤苗,用以治疗行瘤小鼠。应用流式细胞仪(FCM)测定荷瘤小鼠外周血T淋巴细胞亚群的变化,并观察荷瘤小鼠生存期。结果发现,GM-CSF基因转染瘤苗能显著提高小鼠外周血CD8+T淋巴细胞亚群的含量,荷瘤小鼠生存期显著延长。结果表明:GM-CSF基因转染瘤苗能诱导出有效的抗肿瘤免疫反应,提示应用该疗法进行肿瘤基因治疗的可行性。H22 cells, a hepatocellular carcinoma cell line of BALB/C mice origin, were transfected with human GM-CSF via recombinant adenovirus and irradiated by 60Co γray. Bearing mice were treat-ed by above tumor vaccine. The lymphocyte subsets in peripheral blood of bearing mice were determined by Flow Cytometer (FCM). And the survival stage of the animals was also observed. The results showed that the treatment with GM-CSF-tranfected, irradiated H22 cells not only increased the percent-age of CD8+ T-lymphocytes in peripheral blood, but also significantly prolonged the survival stages of the tumor bearing mice. It is concluded that irradiated, GM-CSF-transfected tumor cells can induce ef-fective antitumor immunity, indicating the feasibility of cancer gene therapy.
