摘  要:目的:观察内肥素对THP-1源性泡沫细胞形成中血凝素样氧化低密度脂蛋白受体-1(LOX-1)表达的影响,及其在动脉粥样硬化中的作用机制。方法:给予不同浓度的内肥素处理24h和同一浓度的内肥素处理THP-1源性巨噬细胞0~36h,运用油红O染色观察细胞内脂滴的变化,运用逆转录聚合酶链反应和Western-blot检测LOX-1的mRNA和蛋白的表达水平。结果:高浓度的内肥素处理负荷LDL的THP-1源性巨噬细胞24h后,细胞浆内的脂滴明显增多。在负荷LDL的THP-1源性巨噬细胞上,内肥素上调LOX-1mRNA和蛋白表达。结论:内肥素上调THP-1源性泡沫细胞中LOX-1的表达。Objective:To investigate the impact of visfatin on the expression of Lectin-like oxidized low-density receptor-1(LOX-1) by inducing human monocytic cell line (THP-1)-derived foam cell formation and to evaluate its mechanism of atherosclerosis.Method:THP-1-derived macrophages were incubated with increasing concentrations of visfatin for 24 hours or the same concentration of visfatin from 0 to 36 hours.Lipid droplets in cytoplasm were observed by oil red O staining.The expression of LOX-1 at mRNA and protein levels were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot,respectively.Result:Higher concentrations of visfatin for 24 hours incubation resulted in the large accumulation of lipid droplets in THP-1-derived macrophages when co-cultured with low density lipoprotein (LDL).Visfatin had effection LOX-1 mRNA and protein expression,it up-regulated the expression of LOX-1 mRNA and protein in concentration in THP-1 macrophages when co-cultured with LDL.Conclusion:Visfatin induces THP-1-derived foam cell formation by up-regulating the expression of LOX-1,which may provide a new evidence for the development and progression of atherosclerosis initiated.
