摘  要:[目的]建立小鼠结肠Cajal间质细胞(ICC)的体外培养方法,探讨大黄素对体外培养结肠ICC的毒-效作用。[方法]酶解消化法体外培养KM乳鼠结肠ICC,运用免疫荧光法鉴定结肠ICC,并用光学显微镜观察其生长期的形态变化。应用均匀设计法和MTT比色法探索大黄素不同浓度和作用时间对结肠ICC的影响。[结果]大黄素对结肠ICC最大毒性浓度为1%,最小毒性浓度为0.001%;最大药效浓度为0.000 1%,最小药效浓度为0.000 008%。[结论]成功建立了体外培养KM乳鼠的结肠ICC的方法。大黄素对结肠ICC既有毒性又有药效作用,毒性浓度范围为0.001%~1%;药效浓度范围为0.000 008%~0.000 1%。[Objective]To estabilish the method of culturing interstitial cells of Cajal(ICC)of mice in vitro,and to study the toxic effects of emodin on ICC in vitro.[Methods]ICCs of KM mice's conlon were obtained by enzyme-digestion,being cultured in vitro.Then ICCs were identified with immunofluorescence,of which morphologic changes in proliferative period were observed under light microscopy.With the result above,the methodology of culturing ICC in vitro were fixed.Uniform design methodology and MTT assay were used to study the effect of emodin on ICC in terms of concentration and functional time.[Results]The range of the functional concentration of emodin on ICC in vitro were defined:toxicity range(maximum:1%,minimum:0.001%),efficacy range(maximum:0.000 1%,minimum:0.000 008%).[Conclusion]The method of culturing ICC was established.Emodin has both toxic and therapeutic functions on ICCs in vitro.The toxic concentration of emodin ranges from 0.001% to 1%,while the therapeutic concentration of emodin ranges from 0.000 008% to 0.000 1%.
