葛根素对酒精性肝纤维化影响的实验研究

摘　　要：[目的]研究葛根素对酒精性肝纤维化(HF)的影响。[方法]21只Wistar大鼠随机分为3组,每组7只,对照组用50%葡萄糖20 ml/(kg.d)灌胃,模型组用40%乙醇8 g/(kg.d)灌胃,葛根素组用40%乙醇8 g/(kg.d)灌胃加葛根素10 mg/kg腹腔注射,3组均给药16周。处死大鼠取血清及肝脏,通过光镜观察肝组织病理变化并进行纤维化积分,放免法测定血清透明质酸(HA)、Ⅲ型前胶原(PCⅢ)及层黏连蛋白(LN)。[结果]葛根素明显改善HF程度,与对照组比较差异有统计学意义(P<0.05),与模型组比较,血清HA、PCⅢ及LN显著降低(P<0.05)。[结论]葛根素具有较强的抗HF作用。[Objective]To investigate effect of Peurarin on alcoholic hepatic fibrosis in rats.[Methods]Twenty-one Wistar rats were randomly distributed into three groups at random.The control group received intragastric administration of 50% glucose,20 ml/（kg·d）;The model group received intragastric administration of 40% alcohol,8 g/（kg·d）;The Puerarin group received peritoneal injection of Puerarin,10 mg/kg.All administrations was given continuously for sixteen weeks.Then rats were all executed.Serum and liver tissues were obtained.Serum levels of hyaluronic acid,procollagen Ⅲ and laminin were detected with radioimmunoassays.Pathological changes in liver were observed under light microscope.[Results]Degree of fibrosis was lower in the Puerarin group versus the model group（P0.05）serum levels of hyaluronic acid,procollagen Ⅲand laminin decreased significantly in the Puerarin group versus the model group（P0.05）.[Conclusion]Puerarin has a therapeutic effect on hepatic fibrosis.
