摘  要:目的:探讨脉冲强磁场对体外培养的膀胱肿瘤细胞BIU-87凋亡的影响。方法:将体外培养的膀胱肿瘤细胞株BIU-87随机分为对照组及脉冲强磁场组,脉冲强磁场组置于磁场强度为8 T,重复频率为15次/s的脉冲强磁场中进行干预,每天干预2 h。分别于磁场干预第1天、第2天及第3天后采用倒置显微镜观察细胞生长情况,采用MTT法检测肿瘤细胞凋亡情况(OD值),以流式细胞仪检测膀胱肿瘤细胞BIU-87的凋亡率。结果:倒置显微镜观察脉冲强磁场组细胞于干预第1天后即出现细胞体积变小、核固缩等凋亡表现,磁场干预第3天后可见染色体固缩、碎裂,出现大量脱落的凋亡小体等凋亡。MTT法检测显示脉冲强磁场组干预第1天、第2天及第3天后肿瘤细胞增殖抑制率分别为15.79%、28.84%、37.25%,肿瘤细胞OD值较对照组差异有统计学意义(P<0.05);以流式细胞仪检测脉冲强磁场组肿瘤细胞凋亡率较对照组明显增高,差异有统计学意义(P<0.01)。结论:脉冲强磁场对膀胱肿瘤细胞BIU-87有明显抑制作用,可诱导肿瘤细胞凋亡。Objective: To investigate the effects of high pulsed magnetic field on apoptosis of human bladder cancer BIU-87 cells in vitro. Methods: The human bladder cancer BIU-87 cells were put into 96 hole plates,and randomly divided into high pulsed magnetic field group and control group. High pulsed magnetic field group were exposed to high pulsed magnetic field (8 T,15 Hz) for stimulation 2 hours everyday. Inverted phase contrast microscope was used to observe the cultured cells, MTT assay was applied to reveal viable cells with OD value,and flow cytometry was used to detect the apoptosis result after the first,second and third day. Results: After experi mental treatment,the tumor cells of high pulsed magnetic field group appeared typical apoptotic characteristics including nuclear condensation, chromosome shrinking, nucleus fragments and many apoptotic bodies. The tumor cells proliferation inhibition rate of high pulsed magnetic field group after the first, second and third day were 15.79%,28.84%,37.25%. Compared high pulsed magnetic field group with the control group by MTT assay, the OD value of high pulsed magnetic field group was significantly less than the control group (P〈0.05). The rate of apoptosis cells by flow cytometry was higher in high pulsed magnetic field group than that in control group (P〈0. 01). Conclusions: High pulsed magnetic field conspicuously restrained the growth status of human bladder cancer BIU-87 cells,and the tumor cells could be induced to apoptosis by high pulsed magnetic field.
