摘　　要：基因敲除技术是建立在同源重组技术之上,可对基因组进行定位修饰的实验方法。本文简述了基因敲除技术的基本原理、打靶策略、筛选机制,在动植物和微生物中常用的基因敲除方法以及基因敲除的应用。Gene knockout was built on homologous recombination,and might be positional modified on the genome.This paper introduced the principle,the targeting strategies,the screening mechanism and the commonly method in animals,plants and micro-organisms of gene knockout.The application of gene knockout was also prospected in this paper.
