摘　　要：目的观察SBHL对甲状腺鳞癌SW579细胞生长的抑制作用,从分子水平探讨SBHL对甲状腺鳞癌SW579细胞生长抑制作用的机制。方法用MTT比色法观察不同浓度和作用时间的SBHL对甲状腺鳞癌SW579细胞增殖的影响;用流式细胞术对细胞周期进行分析;TRAP-银染法检测端粒酶活性。结果 5×10-5mol/L-1×10-3mol/LSBHL对甲状腺鳞癌SW579细胞增殖有明显的抑制作用,这种抑制作用随SBHL浓度的增加而增加;流式细胞术对细胞周期进行分析的结果显示,随着SBHL浓度的增加,S期细胞含量逐渐降低,G1期细胞含量逐渐增加,SBHL将细胞阻滞于G1期,抑制DNA合成。SBHL处理的甲状腺鳞癌SW579细胞与对照组相比,端粒酶活性明显降低。结论 (1)SBHL对甲状腺鳞癌SW579细胞生长具有明显的抑制作用,而且这种抑制作用与SBHL的浓度呈正相关。(2)其机制可能与下调端粒酶活性,进而抑制肿瘤细胞生长有关。Objective To observe the inhibiting effects of Sdosodine Hydrochlorida on the growth of thyroid gland cancer SW579 cells and investigate the inhibiting mechanisms from molecular levels.Methods The proliferation of thyroid gland cancer SW579 cells were observed by MTT assay.The changes of the cell cycle of Hela cell treated with Sdosodine Hydrochlorida were analyzed by Flow Cytometry.The tolermerase activity were detected by TRAP-silver staining.Results The proliferation of thyroid gland cancer SW579 cells were inhibited at 1×10^-5mol/L^-1×10^-3mol/L Sdosodine Hydrochlorida.The inhibiting effect increased gradually following the increase of concentration of Sdosodine Hydrochlorida.The percentage of thyroid gland cancer SW579 cells in S phase decreased and the percentage of cells in G1 phase increased after treated by Sdosodine Hydrochloridia,indicating Sdosodine Hydrochlorida arrested thyroid gland cancer SW579 cells in G1 phase and thus the systhesis of DNA was inhibited.After treated by Sdosodine Hydrochlorida,the expressions of telomerase of thyroid gland cancer SW579 cells were downregulated obviously.Conclusion（1）The growth of thyroid gland cancer SW579 cells were inhibited obviously by Sdosodine Hydrochlorida and the inhibiting effect was dependent on concentration of Sdosodine Hydrochlorida;（2）The tolermerase activity were downregulated,the growth of thyroid gland cancer SW579 cells were inhibited by Sdosodine Hydrochlorida.
