摘  要:目的观察银杏达莫注射液对大鼠局灶性脑缺血再灌注损伤中神经细胞Caspase-3蛋白(半胱天冬蛋白酶)表达的影响,探讨其神经保护作用机制。方法将动物随机分为假手术组、生理盐水组及2个药物干预组,参照Zea Longa线栓法建立大鼠左侧大脑中动脉闭塞(MCAO)再灌注模型,给予不同方式处理,在6 h、12 h、24 h 3个时间点分别取材,通过免疫组化的方法检测Caspase-3蛋白的表达变化。结果手术组的大鼠较假手术组Caspase-3蛋白的表达明显增多,其中2个药物干预组Caspase-3蛋白表达较生理盐水对照组明显减少(P<0.01),大剂量银杏达莫组较小剂量银杏达莫组Caspase-3蛋白表达减少(P<0.01)。结论缺血再灌注损伤可导致大鼠脑组织Caspase-3蛋白表达增多,进而导致神经细胞凋亡,银杏达莫注射液可以抑制Caspase-3蛋白表达,减少细胞凋亡数量,从而起到脑保护的作用。Objective It is to observe the influence of the Yinxingdamo on protein expression of Caspase-3 in nerve cells of rat with focal cerebral ischemia-reperfusion injury,to explore its neuroprotective mechanism.Methods The animals were randomly divided into sham-operated group,saline group and two drug interfered groups,the left middle cerebral artery occlusion(MCAO) and reperfusion models in rats were established by Zea Longa suture method,and treated with different sub-manner,the brain tissue were taken at 6 h,12 h,24 h,then the proteinexpression of Caspase-3 were detected by immunohistochemistry.Results The protein expression of Caspase-3 in rats of surgical group was significantly increased than that of sham-operated group,and the protein expression of Caspase-3 in rats of two drugs interfered group were significantly reduced than that of saline group(P〈0.01),the protein expression of Caspase-3 in high-dose Yinxingdamo group was decreased than that in low-dose Yinxingdamo group(P〈0.01).Conclusion Ischemia-reperfusion injury in rat brain tissue can lead to Caspase-3 protein expression increased,leading to neuronal apoptosis,Yinxingdamo injection can inhibit Caspase-3 protein expression,reduce the number of cell apoptosis and thus play a role of protection on cerebral.
