摘 要:目的研究大鼠全脑缺血再灌注损伤后APC-Cdh1的表达变化。方法雄性SD大鼠60只,体重280~350g,随机分成假手术组(SH组)、模型组(IR组)。采用改良4-VO法建立SD大鼠全脑缺血模型,在损伤后不同时间点,免疫组化染色检测Cdh1的表达部位并且采用实时荧光定量PCR检测大鼠海马组织APC-Cdh1的表达。结果与对照组相比,术后1dCdh1mRNA表达减少,术后3d显著升高,术后7d又降低。免疫组化检测显示APC-Cdh1在海马区及皮质区中均有大量表达。结论 APC-Cdh1可能与中枢神经系统的发育及损伤有着密切的关系。Objective To investigate the expression of APC-Cdh1 after cerebral ischemia-reperfusion damage.Methods 60 male Sprague-Dawley rats were randomly divided into Sham-operated group (SH) and model group (IR).The rats of model group were induced by modified four-vessel occlusion (4VO).At different times after injury,the expression of APC-Cdh1 of hippocampus was observed by quantitive real time PCR and examined by immunohistochemistry.Results Compared with control group,the expression of Cdh1 mRNA was significantly decreased on 1d after injury and increased obviously on 3d,but decreased again on 7d.The immuno-staining showed that Cdh1 was highly expressed in cerebral cortex and hippocampus.Conclusion APC-Cdh1 may play an important role in central nervous system development and injury.
