摘  要:目的通过建立二甲基亚硝胺(DMN)诱导大鼠肝纤维化模型,用逆转录聚合酶链反应(RT-PCR)法检测肾素-血管紧张素Ⅱ(AngⅡ)1型受体(AT1R)mRNA表达在大鼠肝纤维化形成过程中的动态变化,观察桃核参术汤对DMN致大鼠肝纤维化形成的干预作用。方法采用DMN诱导的大鼠肝纤维化模型,给予大鼠1%的DMN(1mL/kg体重,1次/d,每周连续3d,共4周)腹腔注射。防治组同时用桃核参术汤干预治疗。模型组和防治组在开始注射DMN后的第14天、第28天、第42天处死动物,取肝组织备用。用RT-PCR法检测AT1RmRNA的表达变化。结果与各模型组比较,防治组大鼠肝组织的AT1RmRNA表达水平均显著降低,与模型组比较,差异有统计学意义(P<0.05或P<0.01)。结论桃核参术汤可以降低DMN大鼠肝组织的AT1RmRNA表达水平。Objective To observe the intervention of taoheshenzhu decoction on liver fibrosis by establishing the models of liver fibrosis in rats induced by dimethylnitrosamine(DMN)and detecting the dynamic changes in renin-angiotensin Ⅱ(AngⅡ)I receptor(AT1R)mRNA expression with reverse transcription-polymerase chain reaction(RT-PCR)during liver fibrosis in rats.Methods The rat model of liver fibrosis induced by DMN was adopted and administered intraperiotoneal injection with 1% DMN(1 mL/kg,1 time /day for 3 consecutive days per week,totally for 4 weeks).Taoheshenshu decoction was applied at same time in prevention group.The animals were sacrificed on the 14th,28th and 42nd days after DMN injection in both model and prevention groups and the liver tissue was collected.RT-PCR assay was used to detect the changes in AT1R mRNA expression.Results Compared with each of model groups,AT1R mRNA expression of rat liver tissue in prevention group decreased significantly,indicating significant statistical difference(P〈0.05 or P〈0.01).Conclusion Taoheshenshu decoction can decrease AT1R mRNA expression in liver tissue with DMN injected in rats.
