摘 要:了解雌激素对大鼠胰腺腺泡细胞癌癌前病变及其K-ras基因突变的影响。方法雄性Wistar大鼠单次腹腔注射重氮乙酰丝氨酸(azaserine)30mg/kg体重,部分大鼠2个月后开始每2周内肌肉注射成酸雌二醇0.18mg/100g体重,4个月后实验结束常规取胰腺组织行光镜和电镜检查,并应用PCR-SSCP法检测K-ras基因突变情况。结果单纯给予Azaserine的大鼠胰腺腺泡细胞癌癌前病变(AACN)发生率100%,K-ras基因突变率42.1%;经雌激素处理后,AACN发生率92.4%,K-ras基因突变率25.6%。结论Azaserine可成功诱导大鼠胰腺腺泡细胞癌癌前病变,K-ras基因突变在此过程中属早期事件。雌激素在一定程度上可抑制该癌癌前病变的发生发展。Objective To set up the animal model of precancerous lesion of rats' pancreatic acinarcellular cancer, learn the effect of estrogen on this lesion's biological feature and K-ras gene mutation.Method Male Wistar rats were injected Azaserine 30 mg/kg (ip). Some rats received estradiol-17 βValerate (0. 18 mg/100 g 2 weeks, im). Four months later, the rats'pancreas was autopsied and K-rasgene mutation was detected by PCR-SSCP method. Result The incidence of AACN was 100%. Theincidence of K-ras gene mutation was 42.1% in rats without estrogen and 25.6% with estrogen. Conclusion Azaserine could induce precancerous lesion of rats' pancreatic acinar cancer successfully. K-rasgene mutation was an early event during this process. Estrogen could inhibit the development of precancerous lesion to some degree.
