摘  要:目的:探讨全反式维甲酸对大鼠C6胶质瘤细胞周期及周期因子的影响及其机制。方法:建立大鼠C6脑胶质瘤模型,腹腔注射全反式维甲酸后,流式细胞仪检测细胞增殖时相变化,免疫组化法检测p27kip1蛋白和Cyc-lin D1蛋白表达的变化。结果:治疗组较对照组,G1期细胞比例增加,S期细胞比例减少,p27kip1蛋白表达增多,Cyc-lin D1蛋白表达减少。结论:维甲酸处理后,大鼠C6胶质瘤细胞周期发生G1期阻滞,其机制同上调p27kip1蛋白表达及下调Cyclin D1蛋白表达有关。Objective To explore the effect of alltrans retinoic acid(ATRA) on cell cycle of rat C6 glioma cells and its possible mechanism.Methods The C6 glioma brain tumour model in rats were prepared and then given ATRA with intraperitoneal injection.The change of cell cycle was determined with flow cytometry,the expression of p27kip1and Cyclin D1 proteins were detected by immunohistochemical staining.Results Compared with control group,the ratio of cells at G1 phase was significantly increased and at S phase was significantly decreased,the expression of p27kip1 protein was upregulated and the expression of Cyclin D1 protein was downregulated in the treatment group.Conclusion The cell cycle of rat C6 glioma cell arrested at G1 phase after treating with ATRA,which may relate to the upregulation of p27kip1 protein and downregulation of Cyclin D1 protein.
