摘  要:目的探讨库普弗细胞在纤维化肝脏中的表达、分布及意义。方法 Wistar大鼠随机分为健康组、肝纤维化模型组,肝纤维化模型组经皮下注射四氯化碳(CCl4)植物油混合液造模成功。苏木素-伊红染色,肝组织羟脯氨酸(Hyp)含量测定,赖氏法测定丙氨酸转氨酶(ALT),TBA法检测丙二醛(MDA)的水平,鲎试剂法测定血浆内毒素水平,免疫组织化学法测定ED2的表达。结果肝脏纤维化组ALT、MDA、Hyp含量明显高于健康对照组(P<0.05)。ET含量呈上升趋势。免疫组织化学结果显示ED2阳性信号在健康对照组沿肝血窦少量散在分布,纤维化模型组阳性信号明显增多(P<0.05),主要分布在增生的纤维组织及纤维间隔,在肝实质内也有少量分布,阳性信号随着实验的进程呈递增趋势,且阳性细胞胞质丰富、体积变大,外形为星形、多角形和不规则形。结论库普弗细胞活化在肝纤维化发生发展过程中有致关重要的作用。Objective To investigate the expression and distribution and significance of kupffer cells (KCs) in experimental hepatic fibrosis. Methods Wistar rats were randomly divided into normal control group and hepatic fibrosis model group. The rats in hepatic fibrisis model group were treated with subcutaneous injection of carboan tetrachloride. The content of hydroxyproline was measured. The alanine aminotransferase (ALT) of plasma was measured by laishi method. The content of malondialdehyde (MDA) and expression of ED2 in liver tissue were determined by TBA and immunohisto-chemistry method, respectively. Results The excertion of ALT, MDA, Hyp and ET in ibrows septuinere mone than group. And the excertion of ET was apgrade with the time. In control group, just a small amount of positive staining of ED2 could be found around fibrous septa. However compared with control group, the expression of ED2 significantly up-regulated mainly in fibrous septa (P〈0.05) and a few hepatic cells. Furthermore, the ED2 positive staining cells take the shape of star, polygon and irregular-shaped and became larger, the cytoplasm of these cells were affluent. Concln^ion KCs activation is the crucial factor during hepatic fibrosis.
