摘　　要：目的探讨FCM技术联合多重PCR技术在慢性淋巴细胞白血病（CLL）基因异常检测的意义。方法收集2008年1月～2010年1月本院CLL患者84例及38例健康正常人的外周血标本和骨髓标本,应用FCM技术联合多重PCR技术进行检验,并注意记录检验结果。结果 CD＋19、CD＋10细胞百分率明显低于正常对照组,CD＋34细胞百分率明显高于正常对照组（P〈0.05）。而性别、年龄阶段不影响CLL患者外周血细胞亚群的表达水平不同性别、不同年龄组之间的比较,性别、年龄阶段不影响CLL患者外周血T细胞亚群表达水平。在检测的84例CLL标本中45例发现异常甲基化,而38例健康正常对照组无1例检测到异常甲基化。结论 FCM技术联合多重PCR技术为肿瘤的诊断、疗效评价和预后提供了重要的参考指标,可提高慢性淋巴细胞白血病的诊断水平,特别是基因异常甲基化的检出率,这结果在慢性淋巴细胞白血病中具有一定的检验价值,能准确地为临床医生选择治疗方案提供依据,提高治疗效果,避免误诊误治,具有明显的临床使用价值。Objective To investigate the FCM technology combined multiple PCR technique in chronic lymphocytic leukemia（CLL） Significance of detection of genetic abnormalities.Methods January 2008 ～ January 2010 84 patients with CLL hospital and 38 healthy normal peripheral blood and bone marrow specimens,the joint application of multiple PCR technique FCM technology inspection,and pay attention to record test results.Results CD ＋19,CD ＋10 cells were obviously lower than the control group,CD ＋34 cells were obviously higher than the control group（P 0.05）.Sex,age does not affect cell subsets in patients with CLL the expression of different sex,comparisons between different age groups,gender,age does not affect the CLL patients the expression of T cell subsets.In the detection of 84 cases of CLL samples,45 cases of abnormal methylation,and 38 healthy normal control group 1 case detected abnormal methylation.Conclusion FCM technology combined multiple PCR technology for cancer diagnosis,prognosis and efficacy evaluation provides an important reference index may improve the diagnosis of chronic lymphocytic leukemia,in particular methylation detection rate,which results in chronic lymphatic leukemia in the value of certain tests,clinicians can accurately provide the basis for selecting treatment options,improve treatment and avoid misdiagnosis and mistreatment,has obvious clinical value.
