摘  要:目的:为了探讨中华眼镜蛇毒(NNAV)对鼻咽癌(NPC)等抑瘤作用。方法:用MTT法检测NNAV对人高、低分化鼻咽癌细胞株(CNE-1,CNE-2)、慢性红白细胞血病(K562)和鼠肝癌(Hep-2)细胞株的体外细胞毒作用;同时在裸鼠异体移植性人CNE-2肿瘤模型观察NNAV对NPC的抑瘤作用。结果:NNAV对4种体外培养的肿瘤细胞均有明显细胞毒效应,NNAV对上述细胞的作用24IC50;分别为79μg/ml,75μg/ml,5.5μg/ml和65μg/ml,其中以红白血病最敏感;NNAV对肿瘤细胞株CNE-1和CNE-2的细胞毒作用呈明显时效关系。NNAV对荷人NPC裸鼠有明显的抑制作用,抑瘤率28.75%-60.13%,呈明显量效关系。结论:在体内外NNAV对NPC等均有抑瘤作用。Objective: To research the inhibitory effect of naja naja actra venom (NNAV) on nasopharyneal carcinoma (NPC) and other tumors in vitro and in vivo. Methods: cytotoxicity effect of NNAV on human cancer cell lines of well and poor differentiated NPC (CNE-1, CNE-2), chronic myelogenous leukemia (K562) and hepatoma (Hep-2) was determined by MTT assay in vitro. Inhibitory effect of NNAV on NPC in BALB/c nude mice (NM) was observed. Results:NNAV has significant cytotoxicity to these four cell lines. The IC50(24 huour incubation) was 79, 75, 5. 5 and 65 μg/ml respectively. Among them K562 was the most sensitive one. cytotoxity effect of NNAV on human cancer cell limes of CNE1 and CNE-2 was in correspondence with time of NNAV. NNAV exhibited significant antitumor activity on nude mice bearing nasopharyngeal cancer. Rates of inhibition were between 28. 75 and 60.13 percent.Inhibitory effect Of NNAV was in correspondence with dose of NNAV. Conclusions:NNAV exhibited inhibitory effect on NPC and other tumors in vitro and in vivo.
