人乳头瘤病毒16与胃癌相关性的研究

摘　　要：目的探讨人乳头瘤病毒１６（ＨＰＶ１６）与胃癌发生的关系。方法用聚合酶链式反应（ＰＣＲ）扩增出ＨＰＶ１６早期区Ｅ６的１２０ｂｐＤＮＡ片段，扩增产物与５＇末端３２Ｐ标记的特异性寡核苷酸探针进行斑点杂交及放射性自显影，用此方法对３４６份新鲜的及福尔马林固定石蜡包埋的胃组织标本中ＨＰＶ１６ＤＮＡ进行检测。结果胃腺癌、癌旁粘膜、胃局部淋巴结及正常胃粘膜组织中ＨＰＶ１６的检出率分别为：２２．７２％（３０／１３２）、５．８８％（４／６８）、０％（０／４２）、２．８８％（３／１０４），胃癌组ＨＰＶ１６的检出率高于其他各组，统计学上差异均有显著性（Ｐ＜０．０１）。结论ＨＰＶ１６可感染胃粘膜上皮细胞，可能是胃癌的致癌因素之一。Objective The study was aimed to clarify the relation of human papillomavirus type 16 (HPV16) with gastric carcinoma.Methods HPV16 DNA in 346 fresh and formalin fixed gastric mucosal specimens embedded in paraffin blocks was assayed with PCR technique. Results HPV 16 DNA was found in 22.72%(30/132)of specimens with gastric adenocarcinoma,in 5.88% (4/68)of mucosa adjacent to malignancy, in 2.88%(3/104)of biopsies with normal mucosa,and none (0/42) of lymph nodes.HPV 16 was more frequently seen in cases with carcinoma than others with significance( P <0 01). Conclusion Infection of HPV 16 in gastric adenoepithelium could be a high risk factor for carcinogenesis.
