摘  要:目的观察褪黑素(MLT)联合全反式维甲酸(AT-RA)对人早幼粒细胞白血病细胞株NB4的增殖、凋亡、分化及细胞周期的影响。方法 1mmol/L的MLT与1μmol/LATRA联合作用于NB4细胞,1mmol/L的MLT和1μmol/LATRA作为对照组,药物作用24、48、72h后进行试验。流式细胞术检测细胞凋亡率、细胞膜表面抗原CD11b的表达阳性率,观察细胞周期的变化情况。结果 MLT联合ATRA作用于NB4细胞24、48、72h后,促进细胞的凋亡率为16·23%±0·38%、31·03%±0·51%和55·65%±2·68%,细胞表面CD11b的阳性率为22·66%±0·36%、57·31%±3·12%和81·74%±0·47%,对细胞周期的影响则表现为G1期细胞增多(68·1%±0·3%、73·3%±0·4%和83·2%±0·2%),S期细胞减少(23·1%±0·1%、23·2%±0·5%和8·2%±0·1%),均优于单用MLT组或ATRA组(P<0·05)。结论 MLT和ATRA联合用药的作用优于单用药。Objective To investigate the effects of melatonin(MLT)and all trans retinoic acid(ATRA)on NB4 cells.Methods The NB4 cells were treated with 1 mmol/L MLT and 1 μmol/L ATRA together for different duration(24,48 and 72 h) in cell culture.The apoptosis,expression of CD11b and cell cycle were measured by flow cytometry.Results After treated with MLT and ATRA together for 24,48 and 72 h,the ratio of apoptotic was 16.23%±0.38%,31.03%±0.51% and 55.65%±2.68%,the expression of CD11b was 22.66%±0.36%,57.31%±3.12% and 81.74%±0.47%, the ratio of G1 phase cells was 68.1%±0.3%,73.3%±0.4% and 83.2%±0.2%,while S phase cells was 23.1%±0.1%,23.2%±0.5% and 8.2%±0.1% respectively.All results were better than those treated with MLT or ATRA alone(P〈0.05).Conclusion Treated with MLT and ATRA together,the increasing of apoptotic or differentiation,and the inhibition of progression from G1 to S phase of the cell cycle are better than those treated with MLT or ATRA alone.
