摘 要:目的观察老鹰茶总黄酮中山奈酚-葡萄糖苷对TGF-β1诱导的肝星状细胞增殖的影响及机制。方法选用SD大鼠,分离出大鼠肝星状细胞(HSC)进行培养;MTT法检测4种黄酮类单体对HSC增殖的抑制作用;RT-PCR方法检测分析其对HSC的Ⅰ型胶原、Ⅲ型胶原、Smad2、Smad3及Smad7受体中mRNA的表达的影响。结果 4种黄酮类单体中山奈酚-葡萄糖苷能够较强的抑制HSC增殖;能够明显下调HSC的Ⅰ型胶原、Ⅲ型胶原、Smad2及Smad3中mR-NA的表达,上调Smad7mRNA的表达。结论老鹰茶总黄酮中山奈酚-葡萄糖苷相比于其它几种单体较明显抑制HSC的增殖作用,对TGF-β/Smad通路的影响可能是其抗肝纤维化的作用机制之一。Objective To investigate the effect and mechanism of kaempferol-glucopyranoside from total flavonoids of Litsea coreana L(TFLC)on the proliferation of hepatic stellate cells induced by TGF-β1.Methods Rat hepatic stellate cells(HSC)which isolated from SD rats were cultured;MTT method for screening of four flavonoid monomers inhibited the proliferation of HSC stronger;Reverse transcription-polymerase chain reaction(RT-PCR)analysis of their HSC was detected in collagenⅠ,collagenⅢ,Smad2,Smad3 and Smad7 mRNA expression.Results In four kinds of flavonoids monomer,kaempferol-glucopyranoside could inhibit HSC proliferation strongly;It could obviously down-regulate collagenⅠ,collagenⅢ,Smad2 and Smad3 mRNA expression,and it could up-regulate Smad7 mRNA expression.Conclusion Compared to several other momomer,kaempferol-glucopyranoside from TFLC is more obvious to inhibit the proliferation of HSC,and the effect of TGF-β-Smad pathway may be a mechanisms of anti-hepatic fibrosis.
