摘  要:目的:根据已发表文献,综合评估尿苷二磷酸葡糖醛基转移酶1A7(UGT1A7)基因多态性与肿瘤易感性之间的关系。方法:按照统一的检索策略在相关中英文数据库中全面检索相关文献,对文献进行筛选、评价后获得相关研究的结果数据,然后在Stata10软件中用Meta分析的方法,计算合并的OR值及95%CI,并进行敏感性分析和发表偏倚的估计。结果:共有国内外17篇合格文献纳入本研究,累计病例和对照数分别为3081和5426例。合并结果显示,携带UGTA7*3等位基因者患肿瘤的危险性是携带野生型UGT1A7*1等位基因型的1.38倍(95%CI=1.18~1.62,P<0.001),携带UGTA7*2等位基因可能与肿瘤的危险性无关。亚组分析显示,UGT1A7*3等位基因型与肝癌和结直肠癌的危险性升高有关。发表偏倚评估未发现明显的偏倚,敏感性分析显示结论稳定可靠。结论:UGT1A7*3等位基因型可能与肿瘤的易感性升高有关。OBJECTIVE:To explore whether the polymorphisms of UDP-glucuronosyltransferase 1A7 (UGT1A7) contribute to the genetic susceptibility to cancer based on the published works.METHODS:All works related to UGT1A7 and cancer risk were searched which were published in English and Chinese and the allele frequencies of UGT1A7 and outcomes were abstracted after assessed under the same criteria.The pooled ORs with their 95%CI were calculated using meta methods in Stata 10 software.Publication bias and sensitivity were evaluated at the same time.RESULTS:Seventeen studies with 3 081 cases and 5 426 controls were included based on the selection criteria.The pooled result indicated UGT1A73 allele could increase the susceptibility to cancer with OR 1.38 (95%CI=1.18-1.62,P0.001) compared to the wild allele UGT1A71,While UGT1A72 allele had no association with cancer risk.Subgroup analysis indicated that UGT1A73 allele was associated with the higher risk of hepatocellular carcinoma and colorectal cancer.The publication bias was not significant and the results were proved credible by sensitivity analysis.CONCLUTION:UGT1A73 allele seems to be involved in the elevated risk of cancer.
