摘 要:目的观察CyclinD1、CyclinE在食管癌和癌周正常食管组织的表达,从量化角度阐明其在食管癌组织中表达的病理学意义。方法常规石蜡包埋、HE切片确诊,用免疫组化MaxVision法检测CyclinD1、CyclinE在食管癌和癌周正常食管组织的表达,Image-proplus图像分析软件对其表达强度进行定量分析,并用表达的阳性单位(positiveunit,PU)反映其表达强度。结果 CyclinD1、CyclinE蛋白在食管癌和正常食管组织中的表达主要定位在细胞核;CyclinD1、CyclinE蛋白在食管鳞状细胞癌和腺鳞癌癌细胞核中表达的PU值高于正常食管组织(P<0.001);CyclinD1、CyclinE蛋白表达的PU值与食管鳞状细胞癌分化程度有关(P<0.001);随着食管鳞状细胞癌分化程度的降低,CyclinD1蛋白表达量逐级增高;CyclinE蛋白在低分化鳞状细胞癌表达的PU值高于高分化及中分化鳞状细胞癌。CyclinD1与CyclinE在食管癌中表达具有相关性(r=0.652,P<0.01)。结论 CyclinD1、CyclinE蛋白的表达与食管癌的发生、发展及食管鳞状细胞癌恶性程度有关,可能成为判断其指标之一。食管癌Cy-clinD1的表达与CyclinE的表达密切相关,CyclinD1和CyclinE可能在食管癌癌变过程中起协同作用。Objective To observe the expressions of CyclinD1 and CyclinE in esophagus carcinoma and the normal esophagus tissue around it and to clarify the significance in esophagus carcinogenesis. Methods CyclinD1 and CyclinE protein expressions were detected by immunohistochemistry (MaxVision method) using the monocional antibody and their intensity were assessed quantitative by Image-- pro plus image analysis system. Results The monoelonal antibody showed specific cell nucleus. CyelinD1 and CyclinE positive unit (PU) were larger in the esophagus squamous carcinoma and the adenosquamous carcinoma than in normal esophagus tissue and there were significant differences (P ~ O. 001). The ex- pression of the protein CyclinD1 in the esophagus squamous carcinoma increased with the decrease of the pathological classification. CyclinE PU of the nuclei was larger in the poorly--differentiated esophagus squamous carcinoma than in the highly--differentiated esophagus squamous carcinoma and the moderate- ly-differentiated esophagus squamous carcinoma and the difference was significant (P ~ 0. 001). There was close correlation between CyclinD1 protein and CyclinE protein. Conclusions The expression of the protein CyclinD1 and CyclinE are related with the generation, development and the degree of esophagus carcinoma. They are considered two valuable markers in evaluating the esophagus carcinoma. CyclinD1 expression is strongly associated with CyclinE expression in esophagus carcinoma.
