摘  要:目的:建立胆囊腺瘤胆汁蛋白质谱,以供筛选胆囊腺瘤的体液标志物。方法:收集胆囊腺瘤及慢性胆囊炎、胆囊结石病人的胆囊胆汁,予脱盐、脱脂等纯化处理,二维色谱-串联质谱分离鉴定肽段,检索SwissProt数据库。结果:胆囊腺瘤病人的胆汁中共鉴定出221种蛋白质,其中2肽及以上匹配蛋白54种,新发现cadherin-3、PHD fingerprotein 20等12种差异表达蛋白。结论:初步建立了胆囊腺瘤胆汁蛋白质谱,新发现的蛋白质可作为胆囊腺瘤的候选标志物,为进一步研究胆囊癌前病变的分子机制奠定了一定基础。Objective To establish a human biliary proteomic table of bile obtained from gallbladder adenoma patients for the purpose of selecting related markers.Methods Human gallbladder bile samples were obtained from gallbladder adenoma and gallstone patients.After removing the bile salts and lipids,peptide fragments were separated by 2D-HPLC,analysed by MS/ MS,and classified by the aid of the SwissProt database.Results A total of 221 proteins were identified from the bile gathered from a gallbladder adenoma patient,of which 54 were identified basing on two or more peptides.Twelve unique proteins such as cadherin-3 and PHD finger protein 20 may be specially useful for the precancerous gallbladder lesion.Conclusions A human biliary proteomic table of gallbladder adenoma is established.Newly discovered proteins may be chosen as candidate markers,which may serve to lay a foundation for further research on cystic carcinogenesis.
