胸水标本满足双向电泳的样本前期处理与制备

摘　　要：目的:建立胸水标本满足双向电泳的样本前期制备方法。方法:采用离心去除细胞成分、去除高丰度白蛋白和IgG、丙酮沉淀去除杂质等前期处理的方法以满足双向电泳的条件。结果:蛋白质的回收率为14.55%;处理前,240μg的胸水蛋白上样量在双向电泳图中可检出(412±24)个蛋白质点,处理后240μg的胸水蛋白上样量可检出(552±20)个蛋白质点。经上述胸水前期处理后得到的双向凝胶电泳图谱更加清晰,检出蛋白质点增加,分辨率提高。结论:成功建立了胸水前期处理满足双向电泳条件的方法,已建立的胸水蛋白双向凝胶电泳技术将为进一步的胸水蛋白质成分研究奠定基础。Objective： To estblish preparation methods of proteome in pleural fluid seperated by using the technique of two-dimensional polyacrylamide gel electrophoresis（2-DE）.Methods： Sample preparations such as removement of cell element,albumin and IgG depletion,and desaltation,were taken to meet the demands of 2-DE.Results： Recovery rate of the preparations was 14.55%.（412±24） protein spots could be detected before albumin depletion,after albumin depletion,and（552±20） protein spots could be detected.A clear 2-DE could be generated from above sample preparations for pleural fluid proteome.Conclusion： Sample preparations for pleural fluid proteome can meet the demands of 2-DE.The established 2-DE technique will benefit advanced research of pleural fluid proteomics.
