摘  要:目的:观察促性腺激素释放激素(LHRH)类似物那法瑞林对体外培养的人乳腺癌细胞BCaP-37生长的影响。方法:培养的BCaP-37细胞,经下列条件作用4d后进行细胞计数:(1)加入10-5~10-9mol/L的那法瑞林;(2)加入10-7~10-10mol/L的雌二醇(E2);(3)在含10-8mol/LE2的条件下,加入10-6~10-8mol/L的那法瑞林或他莫昔芬;(4)加入10ng/ml或50ng/ml的表皮生长因子(EGF);(5)在含10ng/mlEGF的条件下,加入10-5~10-7mol/L的那法瑞林。结果:那法瑞林在10-5~10-9mol/L浓度下对BCaP-37培养细胞的生长无显著抑制作用,在10-6~10-8mol/L浓度下可拮抗10-8mol/LE2对BCaP-37细胞的生长促进作用,对10ng/mlEGF抑制BCaP-37细胞生长的作用无显著影响。他莫昔芬拮抗E2促进BCaP-37生长的作用与那法瑞林相似。结论:那法瑞林对人乳腺癌细胞BCaP-37体外培养生长无显著抑制作用。Objectve: To study the effects of luteinizing hormone releasing hormone (LHRH) analogue nafarelin, estradiol(E 2) and tamoxifen on the growth of BCaP 37 human breast cancer cell line in vitro. Methods: BCaP 37 human breast cancer cells (2.5×10 4/well) were cultured in DMEM with 5% no estradiol bovine serum. The number of BCaP 37 cell was counted after 96 h culture in following conditions respectively: (1) with nafarelin at 10 -5 ~10 -9 mol/L, (2) with E 2 at 10 -7 ~10 -10 mol/L, (3) with nafarelin or tamoxifen at 10 -6 ~10 -8 mol/L stimulated by 10 -8 mol/L E 2, (4) with epidermal growth factor (EGF) at 10 ng/ml or 50 ng/ml, (5) with nafarelin between 10 -5 mol/L and 10 -7 mol/L influenced by EGF at 10 ng/ml. Results: Nafarelin had no inhibitory effect on growth of BCaP 37 cell cultured at 10 -5 ~10 -9 mol/L . At the concentrations between 10 -6 and 10 -8 mol/L, nafarelin or tamoxifen could antagonize the cell growth stimulated by E 2 (10 -8 mol/L) respectively, and nafarelin had no effect on growth of BCaP 37 cell inhibited by EGF (10 ng/ml). Conclusion: Nafarelin has no inhibitory effect on growth of BCaP 37 human breast cancer cell in vitro, but can antagonize the cell growth stimulated by estradiol.
