摘　　要：目的：观察去甲斑蝥素（ＮＣＴＤ）体外对卵巢癌３ＡＯ细胞和ＡＯ细胞的抑制作用，并初步探讨其作用机制。方法：利用细胞培养技术，以结晶紫染色法测定ＮＣＴＤ对卵巢癌３ＡＯ和ＡＯ细胞的抑制作用，并利用流式细胞仪测定分析ＮＣＴＤ作用下两种肿瘤细胞的细胞周期变化。结果：ＮＣＴＤ１μｇ／ｍｌ即对３ＡＯ和ＡＯ细胞有轻微抑制作用，其半数抑制剂量（ＩＣ５０）分别为２０μｇ／ｍｌ和１０μｇ／ｍｌ。ＮＣＴＤ作用６ｈ，３ＡＯ和ＡＯ细胞生长即受到抑制，其抑制率随着药物作用时间延长而升高。在ＮＣＴＤ作用下，３ＡＯ和ＡＯ细胞Ｇ２＋Ｍ期细胞百分比明显升高，而Ｓ期细胞百分比相应地明显降低。结论：ＮＣＴＤ体外对卵巢癌３ＡＯ和ＡＯ细胞有明显抑制作用，其抑制作用呈剂量和时间依赖性。Objective:To evaluate the inhibitory effects of norcantharidine( NCTD) o n ovarian carcinoma 3AO and AO cells cultured in vitro and to reveal its mec hani sm.Methods:Using cell culture technique,the inhibitory effects of NC TD on 3AO an d AO cells were measured by way of crystal violet dyeing.The alterations of cell cycle induced by NCTD were analyzed with flow cytometry.Results:3AO and AO cell s exposed to NCTD 1 μg/ml were inhibited slightly.The IC 50 values of NCTD for the m were 20 μg/ml and 10 μg/ml respectively.The growth of 3AO and AO cel ls was inhibited after being cultured with NCTD for 6 h. The inhibitory rates were increased as time of culture prolonged.The percent age of 3AO and AO cells in G 2+M phase were increased obviously and the percentage of cells in S phase w ere d ecreased accordingly after treated with NCTD. Conclusion: NCTD h as a significant inhibition of ovarian carcinoma 3AO and AO cells,which is in a dose and time depe ndent manner. The mechanism of NCTD induced antiproliferative effect app ears t o involve blockade of tumor cells in the G 2+M phase of the cell cycle.
