摘  要:目的探讨抑癌因子PTEN和血管内皮生长因子(VEGF)蛋白在大肠癌中表达的意义。方法采用免疫组化SP法检测48例大肠癌患者PTEN和VEGF蛋白的表达,探讨其与大肠癌临床病理特征及生物学行为的关系。结果大肠癌中PTEN蛋白表达阳性率为45.8%(22/48),明显低于正常大肠黏膜组织(P〈0.01);VEGF蛋白表达阳性率为72.9%(35/48),明显高于正常大肠黏膜组织的阳性率(P〈0.01);PTEN和VEGF蛋白表达呈负相关(P〈0.01);PTEN低表达和VEGF高表达与大肠癌Dukes分期、浆膜浸润、淋巴结转移有关(均P〈0.01),而与年龄和组织学类型无关(P〉0.05)。结论PTEN和VEGF表达与大肠癌临床病理特征和生物学行为密切相关,联合检测FFEN和VEGF蛋白的表达,对大肠癌的恶性程度及预后判断具有重要的临床意义。Objective To investigate the significance of tumor suppressor gene PTEN and vasular endothelial growth factor (VEGF) protein expressions in colorectal adenocarcinoma. Methods The expressions of PTEN and VEGF protein were examinde with immunohistochemical SP method. Results In colorectal adenocarcinoma, the positive expression rates of PTEN and VEGF proteins were 45.8% (22/48) and 72.9% (35/48), showing the expressions of PTEN and VEGF proteins in cancer tissues were significantly different compared to those in normal mucosa( P 〈 0.01 ). There were not significantly correlated with patients' age or histologica type( P 〉 0.05 ). The expression of PTEN protein was negatively correlated with that of VEGF protein( P 〈0.01 ) , and the low level of PTEN protein expression and the high level of VEGF protein expression were correlated with the Dukes stage, serosa infiltration and lymph node metastasis of colorectal adenocarcinoma( P 〈 0. 01 ). Conclusion The exporessions of PTEN and VEGF protein were signifilcantly correlated with the clinicopathological characteristics and biologic behaviors in colorectal adenocarcinoma, Combined detection of PTEN and VEGF may be helpful to evaluate malignant degree and prognosis of colorectal neoplasms.
