新棘衣棘头虫及黄鳝组织酯酶同工酶分析

摘  要:采用不连续聚丙烯酰胺垂直凝胶电泳分析新棘衣棘头虫和黄鳝的肌肉、肝胰脏、肠道、脾脏、性腺、肾脏及心脏7种组织。电泳结果表明酯酶在黄鳝各组织及新棘衣棘头虫中均有表达,并且具有明显的特异性。肌肉组织仅3条酶带,比其它组织的特异性较明显;肝胰脏组织酯酶活性最强,有12条酶带;脾脏与肾脏组织相似性最高;除肠组织外,其它6个组织均有g酶带,且活性一样强,推测g酶带为黄鳝组织EST酶谱的基础带。肠组织只有2条弱酶带,可能是由于黄鳝的肠组织受到损伤引起。新棘衣棘头虫的两条酶带a和i,这两条酶带在黄鳝的其它组织中也出现,由联合系数和聚类分析发现肠道和棘头虫的酯酶特异性最小,推测黄鳝和新棘衣棘头虫之间可能存在协同进化的关系。Pallisentis celatus and Monopterus albus' seven tissues (muscle,hep atopancreas,intestine,spleen,gonads,kidney and heart) were comparativly analysised by discontinuous polyacrylamide gel electrophoresis.Electrophoresis result showed that esterase were expressed in Monopterus albus' tissues and Pallisentis celatus with obvious specificity.There were only three enzyme bands in muscle,more obvious specificity than other organizations.Esterase activity of the hepatopancreas was the strongest among the tissues,and there were 12 enzyme bands in hepatopancreas.It was the highest similarity between spleen and kidney.Execpt intestinal tissue,the other six organizations had g enzyme band with same activity,it was inferred that g enzyme band was the foundation band of the EST zymogram.Only two weak enzymes bands in intestinal tissue,This might be caused that the intestinal tissue had been injuried.Pallisentis celatus has two bands a and i which also occured in other organizations of M.albus,and the analysis co-factor and cluster showed that between the P.celatus and intestinal of M.albus had the minimum esterase specificity.It was speculated that the coevolution might exist between P.celatus and M.albus.
