摘  要:目的:通过检测EMS1基因在乳腺癌组织中的表达,结合乳腺癌分子分型及各项临床病理参数,分析EMS1 mRNA表达水平在乳腺癌组织检测中的临床意义。方法:采用RT-PCR法检测94例乳腺癌及癌旁组织EMS1mRNA相对含量,以乳腺癌癌旁组织EMS1 mRNA的相对表达量均值为界限,乳腺癌组织分为高表达组和低表达组;采用ELIVISON二步法,对乳腺癌组织进行免疫组化分析并根据基因聚类分析进行分子分型;分析EMS1基因表达与乳腺癌分子分型及各项临床病理参数的关系。结果:乳腺癌组织与癌旁组织中EMS1 mRNA平均表达水平为0.578±0.105和0.397±0.079,高表达率为60.6%(57/94)。Basal-like型乳腺癌中EMS1平均表达水平最高,与其他分子分型比较,差异有统计学意义。EMS1 mRNA高表达与患者年龄≥50岁、绝经后和腋淋巴结转移成正相关,与肿瘤大小、肿瘤分级和TNM分期状况无关。结论:EMS1基因可能是乳腺癌的一种新预后指标。Objective: To detect the expression of EMS1 gene in breast cancer tissues, combining the molecular type and clinicopathological parameters to evaluate the clinical significance of EMS1 expression in breast cancer. Methods: The expression of EMS1 was detected by RT-PCR in surgical neoplastic and paraneoplastic breast tissues from 94 cases of breast cancer patients. The ER,PR, Her2/neu, EGFR status was detected after operation by ELIVISON two-step immunohistochemistry method and according to gene expression clustering analyze breast cancer molecular type, breast cancer tissues were divided into EMS1 mRNA high expression group and low expression group based on the average value of all paraneoplastic breast tissues and the correlation between EMS1 gene expression, the molecular type and clinicopathological parameters of breast cancer was analyzed. Results: The average expression level of the EMS1 mRNA was 0. 578 ±0. 105 and 0. 397 ±0. 079 in all neoplastic and paraneoplastic breast tissues,and 60.6% (57/94) of all neoplastic cases was EMS1 mRNA high expression. EMS1 average expression in the basal-like carcinoma of the breast was the highest than the four other molecular types and had significant difference. The high expression of EMS1 mRNA was positively correlated with age ≥50, postmenopausal status, axillary lymph nodes metastasis. The expression of EMS1 mRNA was independence of tumor size,tumor grade and TNM stage. Conclusion: EMS1 gene may be a new prognosis factor of breast cancer.
