摘  要:目的:观察溃疡性结肠炎(UC)氧化应激及NADPH氧化酶表达的变化特点,探讨双歧杆菌对UC治疗的可能机制。方法:采用葡聚糖硫酸钠(DSS)制作UC小鼠模型,小鼠随机分成3组:正常对照(NC)组、模型(MD)组、双歧杆菌治疗(BT)组。造模同时给予干预治疗7d,停用造模药物后续治疗7d后处死小鼠,结肠组织HE染色;化学比色法测结肠SOD,GPH-Px,MPO和MDA;qRT-PCR测NADPH氧化酶p22phox和gp91phox亚单位mRNA的表达,westonblotting检测其蛋白水平表达并进行统计学分析。结果:MD组结肠SOD,GPH-Px明显低于NC组(P<0.05);MDA、MPO明显高于NC组(P<0.05);BT组结肠SOD,GPH-Px明显高于MD组(P<0.05);MDA、MPO明显低于MD组(P<0.05);MD组结肠组织p22phox和gp91phoxmRNA与蛋白表达明显高于NC组(P<0.05),BT组明显低于MD组(P<0.05)。结论:双歧杆菌抑制UC结肠的氧化应激水平并抑制NADPH氧化酶的表达,减轻了肠道炎症,延缓UC病情的发展、预防复发。Objective To investigate the possible role of bifidobacterium against oxidative stress and NADPH oxidase expression in mice with ulcerative colitis. Methods Mice were randomized to control group (NC), model group (MD), or bifidobacterium group (BT). The model of UC was established by using DSS. MDA, MPO, SOD, and GSH-PX activity were determined by chemical chromatometry. Expressions of P22phox and gp91phox were detected by qRT-PCR and Western blot. Results The activities of MDA and MPO activity were markedly higher but SOD and GSH-Px were lower in group MD than in group NC (P 〈 0.05) ; the activities of MPO and MDA activity were significantly lower but SOD and GSH-Px were higher in group BT than in group MD (P 〈 0.05). The expressions of P22phox and gp91phox were obviously higher in group MD than in group NC (P 〈 0.05), while lower in group BT than in group MD (P 〈 0.05). Conclusions Bifidobacterium inhibits oxidative stress and NADPH oxidase expression in ulcerative colitis, alleviating intestinal inflammation and delaying the development of ulcerative colitis.
