摘 要:目的:探讨沙利度胺(Thal)单药及联合顺铂(cDDP)、5-氟尿嘧啶(5-FU)、依托泊苷(VP-16)对卵巢癌细胞株SKOV3体外增殖的抑制作用。方法:(1)MTT法检测细胞增殖抑制;(2)流式细胞术检测细胞周期;(3)半定量RT-PCR检测血管内皮生长因子(VEGF) mRNA、过氧化物酶体增殖物激活受体γ(PPAR-γ) mRNA的表达。结果:(1)MTT法示Thal单药及联合用药对细胞增殖有抑制作用,且呈浓度和时间依赖性。(2)流式细胞术示Thal能阻滞细胞于G0/G1期,联合用药可增强cDDP、5-FU、VP-16的疗效,显著阻滞细胞于S期。(3)半定量RT-PCR示200、100、50μg/mLThal能抑制VEGF mRNA表达;所有浓度Thal均不影响PPAR-γ mRNA的表达。结论:Thal可抑制卵巢癌细胞株SKOV3体外增殖、阻滞细胞周期、抑制VEGF mRNA表达,联合用药可增强cDDP、5-FU、VP-16的疗效。Objective To investigate the proliferation inhibitory effect of thalidomide and its combinative effect with cisplatin (cDDP),5-fluorouracil (5-FU),etoposide (VP-16) on SKOV3 cells line in ovarian cancer cells. Methods Proliferation inhibitory effect was evaluated by MTT. Cell cycle was determined by flow cytometry. Expression of VEGF mRNA and PPAR-γ mRNA was determined by semiquantitative RT-PCR. Results MTT showed that thalidomide could inhibit proliferation of SKOV3 in vitro as a time-and-dose-dependent manner. Flow cytometry showed that thalidomide could block cell cycle in G0/G1 phase. Combined with cDDP,5-FU,VP-16, thalidomide could reinforce their effect and block cell cycle in S phase significantly. Semiquantitative RT-PCR showed that thalidomide 200,100,50 μg/mL groups could down-regulate the expression of VEGF mRNA. But it didn't influence the expression of PPAR-γ mRNA. Conclusion Thalidomide can inhibit proliferation of SKOV3 in vitro, block cell cycle , and down-regulate the expression of VEGF mRNA. Combine with cDDP, 5-FU, VP- 16, thalidomide can reinforce their effects.
