摘  要:本研究初步探讨转录抑制因子GFI1(growth-factor independence 1)在不同类型白血病中的表达水平及转录抑制因子GFI1表达变化在白血病发病机制中的作用。收集65例初诊白血病患者骨髓细胞标本,其中急性髓系白血病(AML)24例,慢性髓系白血病(CML)18例,急性淋巴细胞白血病(ALL)6例,慢性髓系白血病急变期患者17例。缺铁性贫血患者13例作为对照组。采用半定量和实时定量RT-PCR方法检测gfi1的转录表达水平。结果表明:白血病患者组gfi1表达明显高于对照组,差异具有统计学意义(p<0.01),其中初诊CML组gfi1表达显著高于初诊AML组、初诊ALL组及CML急变组,差异具有统计学意义(p<0.01)。CML急变组中,急淋变组gfi1表达明显高于非急淋变组,两组间有显著性差异(p=0.004)。结论:gfi1表达异常可能参与白血病的发生与发展,gfi1表达上调可能促进淋巴系肿瘤的发生。This study was purposed to investigate the expression of the growth-factor independence 1 ( GFI1 ) in patients with leukemia and its clinical significance. Bone marrow mononuclear cells were obtained from 65 newly diagnosed leukemia patients including 24 acute myeloid leukemia( AML), 18 chronic myelogenous leukemia (CML) ,6 acute lymphoblastic leukemia (ALL), 17 blast crisis of chronic myelogenous leukemia and 13 patients with iron deficiency anemia(IDA) were used as controls. The relative expression of gene gill was detected by reverse transcriptase-polyme- rase chain reaction (RT-PCR) and taqman quantitative real-time reverse transcription polymerase chain reaction (QRT- PCR). The results showed that gene expression of gene gill in leukemia patients was obviously higher than that in controis and the difference was statistically significant(p 〈 0.01 ), in which the expression of gene gill in newly diagnosed CML patients was higher than that in newly diagnosed AML, newly diagnosed ALL, CML-BCP patients and the diffe- rence was significant (p 〈0. 01 ). Expression of gene gill in lymphocytic blast crisis of CML was higher than that in nonlymphocytic blast crisis of CML, and the difference was significant. It is concluded that gene gill may play an im- portant role in leukemia, especically in CML incidence and progression. The high level expression of gene gfil may be participate in the development of lymphoma.
