摘 要:目的 :睾丸酮 (T) /乳酸 -乙醇酸共聚物 (PLGA)微球6 0 Co辐照灭菌可行性研究。方法 :T/PLGA微球在6 0 Co源下 ,以 2 5kGy的剂量进行辐照 ,对微球辐照前后主药T、载体PLGA的理化特性和微球体外释药速率的变化进行比较 ,考察6 0 Co辐照对T/PLGA微球理化特性及药物体外释放的影响。结果 :6 0 Co辐照引起PLGA分子量一定程度的下降 ,但对T/PLGA微球的理化特性及体外释药速率没有影响。结论 :初步的可行性研究表明T/PLGA微球可用6 0OBJECTIVE:To investigate the possibility to sterilize testosterone(T)/poly(DL-lactide-co-glycolide)(PLGA) microspheres with 60 Co irradiation.METHOD:The microspheres were irradiated by γ-rays 25kGy using cobalt-60 as the irradiative source.The changes in the physicochemical characteristics of T in microspheres were observed by melting point,thin-layer chromatography and proton nuclear magnetic resonance.The content of T in the microspheres and the release of T from the microspheres were tested before and after the irradiation.The changes in intrinsic viscosity of PLGA were determined using an ubbelodhe viscometer at 25℃ with chloroform as a solvent.RESULTS:The irradiation didn′t have any effect on the physicochemical properties of T and had no significant effect on the drug release profile in vitro though a little decrease in molecular weight of PLGA was caused by the irradiation.CONCLUSION:It is possible to sterilize T/PLGA microspheres with 60 Co irradiation.
