摘 要:目的观察地龙注射液对细胞哮喘模型中转化生长因子(TGF)-β1、Smad2表达的影响,探讨地龙在支气管哮喘治疗中的作用机制及价值。方法给予人支气管上皮细胞系BEP2D(HBE)以不同的干预和治疗,分为:单纯培养组即对照组(A组),屋尘螨(dermatophagoides pteronyssinus,Derp1)(10μg.mL-1)组(B组),Derp1(10μg.mL-1)+地塞米松(dexametha-sone,DEX)(1×10-6mol.L-1)组(C组),Derp1(10μg.mL-1)+地龙注射液(0.9mg.mL-1)组(D组),Derp1(10μg.mL-1)+地龙注射液(9mg.mL-1)组(E组),Derp1(10μg.mL-1)+地龙注射液(18mg.mL-1)组(F组),培养72h,RT-PCR法检测各组细胞TGF-β1mRNA,Smad2mRNA表达;并收集以上各组上清液,作用于人胚肺成纤维细胞(HLF)72h,免疫细胞化学法检测纤维连接蛋白(Fibronectin,FN)表达。结果 B组TGF-β1mRNA,Smad2mRNA表达较A组明显升高(P<0.01);各地龙组及C组,TGF-β1mRNA、Smad2mRNA表达低于B组,有显著性差异(P<0.01),与A组比较均无显著性差异(P>0.05);各组HLF中FN表达与HBE各对应组TGF-β1mRNA表达呈正相关性。结论地龙注射液有抑制哮喘气道重构的作用,其机制可能与抑制TGF-β1/Smad2信号通路有关。OBJECTIVE To observe the effects of Earthworm injection on the human bronchial epithelial cells(HBE) sensitized by house dust mite allergen Dermatophagoides pteronyssinus and human embryonic lung fibroblast(HLF),and to investigate the mechanism on the treatment of asthma.METHODS Using different intervention and treatment,HBE were divided into 6 groups,control group(group A),Derp1 allergen(10 μg·mL-1)group(group B),Derp1 allergen(10 μg·mL-1)+ Dexamethasone(DEX)(1×10-6 mol·L-1) group (group C),Derp1 allergen(10 μg·mL-1)+ Earthworm iniection(0.9 mg·mL-1)group(group D),Derp1 allergen(10 μg·mL-1)+ Earthworm injection(9 mg·mL-1)group(group E),Derp1 allergen(10 μg·mL-1)+ Earthworm injection(18 mg·mL-1)group(group F),cultured for 72 h.Then the expression of transforming growth factors of (TGF)-β1(TGF-β1) mRNA and Smad2 mRNA were determined by RT-PCR.The supernatants of each group were collected and inculoated with the HLF for 72 h.The expression of fibronectin(FN) was detected by immunocytochemical method.RESULTS Compared with the group A,TGF-β1 mRNA and Smad2 mRNA in group B were increased significantly(P0.01).The TGF-β1 mRNA and Smad2 mRNA in group D,group E,group F and group C were all decreased compared with group B(P0.01) and no significant difference from group A(P0.05).After inculoating with HLF for 72 h.The expression of FN was relative with the change of TGF-β1 mRNA,in positive correlation.CONCLUSION The earthworm injection had an inhibitory action on airway remodeling of asthma.It significantly depressed the signaling pathway of TGF-β1/Smad2 and may be a role in the modulating mechanism of asthma.
