摘  要:目的:分析研究抗肺癌24H7单抗的体内外功能,并初步鉴定其识别的抗原蛋白,为肺癌靶向治疗提供候选抗体药物及靶标。方法:采用活细胞免疫荧光及流式细胞荧光检测单抗与多种肺癌细胞的反应;应用CCK-8法和裸鼠体内成瘤实验鉴定单抗对肺癌细胞体外增殖及体内肿瘤生长的影响;同时采用免疫组化检测该单抗识别抗原在肿瘤及癌旁正常组织中的表达差异;免疫亲和层析纯化抗原。结果:活细胞免疫荧光及流式荧光结果显示24H7单抗识别抗原在12种不同肺癌细胞系膜上均有阳性表达;体外增殖及动物体内治疗实验显示该单抗在体外能抑制肺癌细胞的增殖,并在体内显著抑制肿瘤的生长,其抑制率达73.74%(P<0.01);免疫组化结果表明该单抗的靶抗原在肺腺癌组织中的表达较癌旁正常显著上调;免疫亲和层析显示靶抗原的分子量约为170 KD。结论:抗肺癌单抗24H7能够在体内外抑制肺癌的生长,其靶抗原可表达于肺癌细胞的膜上,且在肺腺癌组织上调表达,可能是一个肺腺癌靶向治疗的新靶标。Objective:Study the function in vitro and in vivo of 24H7 monoclonal antibody targeting lung cancer and identify the antigen,to provide a candidate antibody drug and molecule target for the anti-lung cancer therapy.Methods:Binding with viable lung cancer cells of 24H7 monoclonal antibody were studied by immunofluorescence and flow cytometry of viable cells;the inhibition of lung cancer cells and implanted tumor of 24H7 antibody were studied by cell proliferation assay and tumor treatment experiment in vivo;the expressional difference of antigen in lung cancer tissues and corresponding normal tissues of 24H7 antibody was detected by immunohistochemistry;the antigen of 24H7 antibody were purified by immunoaffinity chromatography.Results:The 24H7 antibody could recognize cell membrane protein of 12 lung cancer cell lines;obviously inhibit lung cancer cells proliferation and tumor growth in vivo by 73.74%(P0.01).Immunohistochemistry showed higher expression on the adenocarcinoma of lung cancer tissues.The antigen of 24H7 antibody which molecular weight about 170 KD gained by immunoaffinity chromatography.Conclusion:24H7 monoclonal antibody can surpress lung cancer cells proliferation in vitro and tumor growth in vivo;the antigen of 24H7 expressed on cell membrane and highly on the adenocarcinoma of lung cancer tissues,it might become a candidate molecular for targeted adenocarcinoma of lung cancer treatment.
