急性肝坏死动物模型肠道SIgA的变化

摘　　要：目的研究急性肝坏死动物模型肠道SIgA、IgA的变化,以了解急性肝坏死肠道免疫屏障的变化。方法用ELISA方法、放射免疫方法检测血清和粪便中的IgA、SIgA;采用免疫组化检测急性肝坏死动物模型肠组织IgA的变化。结果肝坏死小鼠便SIgA、IgA含量升高,与生理盐水对照组相比均有统计学差异(P<0.01),在9h达到高峰,但急性肝坏死动物模型肠组织IgA染色较正常肠组织明显减弱,免疫组化光密度分析显示急性肝坏死肠组织IgA较生理盐水对照组明显降低(P<0.01)。结论急性肝坏死肠上皮IgA(SIgA)减少导致肠道免疫屏障损伤,引起肠道免疫防御功能减弱,导致肠道屏障功能发生异常,是引起腹部症状的原由之一。Objective To study the change of SIgA of gut in acute hepatic necrosis and to understand the change of intestinal immunology barrier in the acute hepatic necrosis.Methods We made full use of ELISA,radioimmunoassay to detect IgA,SIgA of blood and feces in the acute hepatic necrosis.As well as IgA of intestinal tissue in the acute hepatic necrosis was detected by immunohistochemistry.Results IgA of serum and feces as well as fecal SIgA in the acute hepatic necrosis were remarkably increased compared with the control（P〈0.01）.But IgA of intestinal tissue was remarkably decreased.The IgA was notably decreased compared with normal control through immunohistochemistry photodensitometry（P〈0.01）.Conclusion The IgA（SIgA） of intestinal epithelium decrement in acute hepatic necrosis induced falling of intestinal immunology barrier and the attenauation of intestinal immunology defence.This is one of the reasons which results the abnormal symptom in acute hepatic necrosis.
