摘  要:采用水提醇沉方法提取玉竹(Polygonatum odoratum(Mill)Druce)粗多糖,通过DEAE-纤维素离子交换层析和Sepharose CL-6B分子筛层析分离纯化得到玉竹中性多糖。应用紫外光谱分析中性多糖的纯度,凝胶过滤法测定其分子质量,红外光谱和高效液相色谱对其结构和单糖组成进行初步分析。结果表明:玉竹多糖主要为中性多糖,同时含有少量酸性多糖;玉竹中性多糖平均分子质量为1.21×106D,主要由甘露糖和葡萄糖组成,其物质的量比为5:1,同时含有少量半乳糖。Crude polysaccharides were obtained from Polygonatum odoratum(Mill) Druce by water extraction and ethanol precipitation,and were sequentially purified by ion exchange chromatography on DEAE-cellulose column and gel permeation chromatography on Sepharose CL-6B column and a neutral polysaccharide fraction was obtained.Its structure was determined using HPLC,FTIR and UV spectroscopy.The results showed that the polysaccharide fraction was mainly composed of mannose and glucose at a molar ratio of 5:1 and also contained a small amount of galactose and that its molecular weight was approximately 1.21 × 106 D.
