摘 要:本试验旨在研究添加不同剂量的沙棘叶总黄酮对1~21日龄爱拔益加(AA)肉鸡蛋白质利用率的影响。选取240只1日龄AA肉鸡,单因素随机分为4组。对照组饲喂基础饲粮,试验组饲喂在基础饲粮中分别添加0.025%、0.050%和0.100%沙棘叶总黄酮的饲粮。饲养试验时间为21d。结果表明,与对照组相比,0.025%和0.050%沙棘叶总黄酮组蛋白质表观消化率分别显著提高了19.43%(P<0.05)和19.29%(P<0.05)。0.025%和0.100%沙棘叶总黄酮组与对照组相比,胸肌率分别显著提高了26.95%和17.42%(P<0.05),腿肌率分别显著提高了37.20%和21.28%(P<0.05)。结果提示,沙棘叶总黄酮可以促进1~21日龄AA肉鸡蛋白质表观消化率和蛋白质的沉积。十二指肠组织蛋白检测结果显示,沙棘叶总黄酮可能通过上调核糖体蛋白12(RPS12)、肠脂肪酸结合蛋白(I-FABP)和角蛋白10(CK10)的表达,促进肉鸡幼雏对蛋白质的消化吸收,从而提高了1~21日龄肉鸡对饲料中蛋白质的利用率。由此说明添加沙棘叶总黄酮可以通过上调十二指肠RPS12、I-FABP和CK10的表达提高1~21日龄AA肉鸡生长前期蛋白质表观消化率,促进胸肌和腿肌的生长发育。The experiment was conducted to evaluate the effects of total flavonoids of sea buckthorn leaves on protein utilization in AA broilers.Two hundred and forty one-day-old AA broilers were randomly allotted into the control group and three experimental groups.The control group was fed the basal diet and the experimental groups were fed the basal diets added with 0.025%,0.050% and 0.100% total flavonoids,respectively.The experiment lasted for 21 days.The results showed that 0.025% and 0.050% total flavonoids of sea buckthorn leaves significantly improved the apparent digestibility of protein(ADP)by 19.43%(P0.05)and 19.29%(P0.05),0.025% and 0.100% total flavonoids of sea buckthorn leaves significantly increased the breast muscle ratio by 26.95% and 17.42%(P0.05),and increased the thigh muscle ratio by 37.20% and 21.28% in AA broilers(P0.05),respectively.The results indicated that total flavonoids of sea buckthorn leaves improved the apparent digestibility of protein and protein deposition in AA broilers.The results of differential expression proteins of duodenal tissue by MALDI-TOF-MS showed that ribosomal protein S12(RPS12),fatty acid binding protein(I-FABP)and cytokeratin 10(CK10),which significantly improved the absorption of protein and resulted in the growth of protein utilization in AA broilers,were significantly increased comparing with the control group.In conclusion,total flavonoids of sea buckthorn leaves could improve the apparent digestibility of protein and the growth of breast muscle and thigh muscle by up-regulating the expression of RPS12,I-FABP and CK10.
