摘  要:目的观察血红素氧合酶-1(HO-1)在慢性非细菌性前列腺炎(CNP)大鼠模型中的表达,探讨能否通过其激动剂氯高铁血红素(Hemin)激活防治前列腺炎以及其可能的发生机制。方法 45只SD大鼠分为正常对照组(对照组)、前列腺炎模型组(模型组)和前列腺炎模型+氯高铁血红素治疗组(治疗组),每组15只。RT-PCR对各组前列腺组织HO-1进行定量分析,免疫组化法检测NF-κB,I κBα,TNF-α的表达变化。结果 (1)HO-l mRNA在模型组中相对表达量明显低于对照组和治疗组(P<0.05)。(2)Hemin治疗组炎症反应减轻,NF-κ B和TNF-α表达明显下降(P<0.05),I κ B α明显增高(P<0.05)。结论 HO-1参与了前列腺炎的发病过程,激动剂Hemin可有效减轻炎症程度,其机制可能为抑制NF-κ B信号通路阻断炎症细胞因子表达和CNP的治疗提供了新的思路。Objective To investigate the expression of Heme oxygenase-1(HO-1) in the rat model with nonbacterial chronic prostatitis and explore its possible meachnism. Methods Total of 45 SpragueDawley rats were randomly divided into the control group(15 rats), the rat model group with nonbacterial chronic prostatitis (15 rats) and the treated rat model group with Hemin (15 rats). The expression of HO-1 in rat prostatic tissue was detected by RT-PCR., and the expressions ofNF- KB, IKBα and TNF- a were detected by immunohistochemistry. Results (1) The expresssion of HO-1 in the rat model group was significantly lower than that of the control group and the treated rat group (P〈0.05). (2) Compared with those in the rat model group, the expressions of NF- K B and TNF- α in the treated rat model obviously decreased (P〈0.05), whereas the expression of I K B α significantly increased (P〈0.05). Conclusion HO-1 might be involved in the pathological development of nonbacterial chronic prostatitis. HO-1 ligands Hemin could ameliorate pathological symptom of nonbacterial chronic prostatitis.
