摘  要:目的研究半边旗中二萜类有效成分之一6F对肺癌细胞细胞周期及细胞中DNA、RNA和蛋白质合成的影响。方法采用体外培养的人癌细胞株——肺腺癌上皮细胞SPC-A-1作实验对象,以噻唑蓝(MTT)法测定细胞存活率;用细胞光度术(FCM)测定6F对SPC-A-1细胞周期的影响[3H]-TdR、[3H]-UTR和[3H]-Leu参入的方法观察6F对SPC-A-1细胞DNA、RNA和蛋白质合成的影响。结果化合物6F可强烈抑制SPC-A-1细胞的生长,24h的半数抑制浓度IC50为2.34μmol·L-1;细胞周期分析显示6F作用细胞24h后,均能使细胞的G2/M期细胞比例升高,同时G0/G1期比例下降,并呈一定的剂量效应关系,6F对[3H]-TdR、[3H]-UTP和[3H]-Leu参入作用在培养24h后均有明显抑制作用,且呈剂量依赖关系。结论化合物6F明显抑制SPC-A-1细胞的生长,阻断细胞于G2/M期,抑制细胞DNA、RNA和蛋白质的合成。AIM To study the effects of compound 6F, one of the diterpenoids from Pteris Semipinnata L on cell cycle and the synthesis of DNA, RNA and protein of lung adenocarcinoma cell line SPC A 1. METHODS Cell viability was determined by using 3 (4,5 Dimethyl 2 thiazolyl) 2,5 diphenyl 2H tetrazolium bromide (MTT) test, the cell cycle was analysed by flow cytometry (FCM) and the synthesis of DNA, RNA and protein was observed by the TdR、 UTP and Leu incorporation. RESULTS Compound 6F strongly inhibited the growth of SPC A 1 cell, the IC 50 value was 2 34 μmol·L -1 after 24 h treatment. FCM analysis revealed that 6F could increase the percentage of cells in G 2/M phase and decrease those in G 0/G 1 phase after 24 h treatment, and in a dose dependent manner, while those in S phase changed a little. The assays of TdR、 UTP and Leu incorporation into DNA, RNA and protein respectively showed obvious inhibition dose dependently after exposure to 6F for 24 h. CONCLUSION Compound 6F remarkbly inhibited SPC A 1 cell line proliferation, arrested SPC A 1 cell in G 2/M phase and inhibited the synthesis of DNA, RNA and protein.
