摘　　要：目的：研究白细胞介素１０对白细胞介素诱导的人肾小球系膜细胞的细胞间粘附分子１表达的影响及其细胞内作用机制。方法：用细胞ＥＬＩＳＡ法检测，ＣＤ５４蛋白水平表达，以Ｎｏｒｔｈｅｒｎ杂交方法测定ＣＤ５４的基因表达，采用电泳迁移率变动法观察核因子ｋＢ的活性。To explore the effect of rhIL-10 on rhIL-1β-induced intercellular adhesion molecule 1 （CD54 ） expression in human mesangial cells and its relation with nuclear factor κB. Methods: Cell ELISA, Northern blot and electrophoresis mobility shift assay were used to determine the protein and mRNA levels of CD54 and the activity of nuclear factor κB in cultured human mesangial cells. Results: rhIL-1β- induced CD54 protein expression in human mesangial cells was inhibited by 20 ng· L-1 rhIL-10. The inhibitory effect of rhIL-10 could last 36 hours after rhIL-1β was added. rhIL-10 could also inhibit CD54 gene expression and nuclear factor κB activity induced by rhIL-1β. Conclusion: rhIL-10 can inhibit rhIL-1β-induced CD54 expression in human mesangial cells and the inhibitory effect may be mediated, at least in part, by a decreased activity of nuclear factor κB.
