摘  要:目的观察siRNA抑制生存素survivin基因对裸鼠皮下人腺样囊性癌ACC-2细胞移植瘤血管生成的影响。方法裸鼠15只完全随机分为3组(n=5):①干扰质粒组,皮下接种survivinRNA干扰质粒转染的ACC-2细胞;②瘤体注射组,皮下接种正常ACC-2细胞15d成瘤后,将针对survivin的siRNA真核表达载体脂质体法作瘤体内及瘤周注射;③肿瘤细胞组,皮下接种正常ACC-2细胞。25d后处死全部裸鼠,剥离瘤体,测量瘤体体积,RT-PCR检测瘤体组织VEGFmRNA表达,HE染色和免疫组化染色法观察VEGF阳性染色及其微血管参数。结果干扰质粒组和瘤体注射组VEGF电泳条带的相对光密度比值(0.698±0.023、0.612±0.038)及总体微血管密度[(4.98±1.43)、(5.08±1.29)个]、有腔微血管密度[(2.98±1.05)、(3.10±1.23)个]、有腔血管周长[(68.60±15.98)、(70.12±14.19)μm]及管腔面积[(4119.14±698.12)、(4339.67±599.76)μm2]较肿瘤细胞组VEGF电泳条带的相对光密度比值(0.912±0.039)及总体微血管密度[(8.90±1.36)个]、有腔微血管密度[(5.89±1.68)个]、有腔血管周长[(129.48±25.68)μm]及管腔面积[(6952.74±1298.54)μm2]低(P<0.05),而干扰质粒组和瘤体注射组无明显差别(P>0.05)。结论 survivinsiRNA能够抑制腺样囊性癌瘤体生长,并在体内通过抑制腺样囊性癌VEGF表达有效减少肿瘤血管生成。Objective To observe the inhibitory effect of silencing survivin through siRNA on the tumor angiogenesis in nude mice bearing human adenoid cystic carcinoma ACC-2 cells.Methods A total of 15 nude mice were randomized into 3 groups (n=5),intervention plasmid group (inoculated subcutaneously with survivin RNAi plasmid transfected ACC-2 cells),tumor intervention group (survivin RNAi plasmids were subcutaneously injected into and around the tumor mass which were formed in 15 d after ACC-2 cells inoculation into the nude mice),and tumor cell group (inoculated subcutaneously with wide ACC-2 cells).After all mice were sacrificed in 25 d after tumor cell inoculation,the tumor was stripped,and tumor volume was measured.RT-PCR was used to detect the expression of VEGF mRNA in tumor tissues,and HE staining and immunohistochemical staining were preformed for the morphology and microvessel parameters of the tumor mass.Results In the intervention plasmid group and tumor intervention group,VEGF mRNA was expressed at an optical density ratio of 0.698±0.023 and 0.612±0.038 in electrophoregram,overall microvessel density was 4.98±1.40 and 5.08±1.29,and a cavity microvascular density was 2.98±1.05 and 3.10±1.23 with an average circumference of blood vessel lumen 68.60±15.98 and 70.12±14.19 μm,and a mean lumen area of 4 119.14± 698.12 and 4 339.67±599.76 μm2.These indexes had a significant difference with those of tumor cell group [optical density ratio of VEGF 0.912±0.039,overall microvessel density 8.90±1.36,a cavity microvessel density 5.89±1.68,an average circumference of vascular cavity 129.48±25.68 μm and lumen area 6 952.74±1 298.54 μm2 (P0.05)],while,there was no significant difference in these indexes between the intervention plasmid group and tumor intervention group (P0.05).Conclusion Survivin siRNA inhibits the growth of tumor mass in adenoid cystic carcinoma-bearing nude mice by inhibiting VEGF expression to suppress tumor angiogenesis effectively.
