摘　　要：目的检测Piwil2在正常宫颈、宫颈上皮内瘤变和宫颈癌中的表达,同时检测HeLa细胞株诱导分化治疗前后Piwil2的表达情况,进而探讨Piwil2在宫颈癌临床诊断中的意义。方法采用Western-blot和免疫组织化学SP法检测正常宫颈、宫颈上皮内瘤变和宫颈癌中Piwil2的表达差异;以及HeLa细胞诱导分化前后Piwil2的表达差异,并结合临床资料分析。结果 Piwil2在宫颈癌和宫颈上皮内瘤变的阳性表达率显著高于正常宫颈组织,差异有统计学意义(P<0.05)。HeLa细胞经诱导分化治疗,Piwil2表达与对照组比较明显降低。结论 Piwil2在CIN及宫颈癌组织中的表达异常可作为宫颈癌及癌前病变的一项辅助诊断指标,也可以作为一项疗效评价指标。Objective To investigate the expressions of Piwil2 in normal cervix,cervical intraepithelial neoplasia,cervical cancer and Hela cell before and after treatment,and then discuss the clinical significance of Piwil2 in cervical cancer. Methods Western blot and immunocytochemistry methods were used to examine the expression levels of Piwil2 in normal cervix,cervical intraepithelial neoplasia,cervical cancer and Hela cells before and after treatment. Results The positive expression of Piwil2 were detected in a significantly greater proportion of cervical cancer and cervical intraepithelial neoplasia than normal cervix （ P〈 0. 05） ,and bring down obviously in Hela cells after treatment. Conclusion Piwil2 expression in cervical intraepithelial neoplasia and cervical cancer could be used as a tumor marker for early diagnosis of cervical cancer and precancerous lesions,and also could be used as an index for therapeutic effect.
