摘  要:目的 探讨p16/p38 MAPK/p53/Wip1通路在乳腺癌发生、发展中的作用及其临床意义.方法 应用免疫组织化学方法检测70例乳腺癌组织、癌旁组织、20例正常乳腺组织中Wip1、p53、p38 MAPK、p16蛋白的表达,并对Wip1蛋白高表达与p53、p38、p16蛋白表达进行相关分析.结果 3种组织中Wip1蛋白高表达率分别为62.9%(44/70)、2.9%(2/70)、0(0/20).乳腺癌组织比癌旁组织、正常乳腺组织明显升高(P<0.01).Wip1蛋白高表达与p53、p38、p16蛋白表达呈负相关(P<0.01,等级相关系数rs分别为-0.529、-0.626、-0.499).结论 p16/p38 MAPK/p53/Wip1是负反馈通路,它可能在乳腺癌发生发展中起重要作用.Objective To investigate the role and clinical significance of the pl6/p38 MAPK/ p53/Wipl pathway in the pathogenesis and development of breast cancer. Methods Immunohistochemis-try was used to detect the expression of Wipl, p53, p38 MAPK and pl6 protein in 70 breast cancer tissues, corresponding noncancerous tissues and 20 normal breast tissues. The correlation between the Wipl protein overexpression and p53, p38, pl6 protein expression was analyzed. Results Wipl protein was overexpressed in 63.9% (44/70), 3.9% (2/70), 0 (0/20) of the breast cancer, corresponding noncancerous tissues and normal breast tissues, respectively. The Wipl protein expression was significantly up-regulated in the breast cancer tissue as compared with that in the corresponding noncancerous tissues and normal breast tissues ( P 〈 0.01). Overexpression of Wipl protein was inversely correlated with the expression of p53, p38, pl6 protein (P 〈 0.01, rank correlation coefficient r, was - 0.529, - 0.626, - 0.499 respectively). Conclusion The pl6/p38 MAPK/p53/Wipl pathway is a negative feedback pathway, which may play a key role in the pathogenesis and development of breast cancer.
