胰腺癌细胞株化疗敏感性与胸苷酸合成酶蛋白表达的关系

摘　　要：目的 探讨胸苷酸合成酶（TS）在胰腺癌细胞中的表达及其蛋白表达水平与胰腺癌细胞化疗敏感性的关系.方法 利用CCK-8试剂盒检测7株胰腺癌细胞（AsPC-1、BxPC-3、Capan-1、SU86.86、T3M4、PANC-1与COLO357）对5-氟脲嘧啶（5-Fu）的化疗敏感性,应用Western blot检测其TS蛋白表达.结果 细胞株AsPC-1、BxPC-3、Capan-1、SU86.86、T3M4中TS蛋白表达水平低（低TS表达组）,而PANC-1与COLO357中TS蛋白表达水平较高（高TS表达组）,低TS表达组的细胞在低中高3个不同药物浓度下对5-Fu抑制率分别为0.683±0.131、0.769±0.114与0.836±0.094.同浓度下高TS表达组的细胞对5-Fu抑制率分别为0.370±0.157、0.548±0.018与0.638±0.020.TS蛋白表达低的细胞对5-Fu化疗敏感性明显高于TS表达高的细胞,差异有统计学意义（P＜0.05）.结论 在胰腺癌细胞株中,TS蛋白表达水平与其对5-Fu的化疗敏感性密切相关.Objective To investigate the relationship between the thymidylate synthase （TS） protein expression levels and chemosensitivity of different pancreatic cancer cells to 5-Fu. Methods The chemosensitivity of seven pancreatic carcinoma cell lines to 5-Fu in vitro was evaluated with CCK-8 kit. TS protein expression of pancreatic carcinoma cells was detected by Western blotting. Results Pancreatic carcinoma cells AsPC-1, BxPC-3, Capan-1, SU86.86 and T3M4 expressed low amounts of TS protein （low TS protein group）. Cells of PANC-1 and COLO357 expressed high amounts of TS protein （high TS protein group） in seven cell lines. The inhibition rate （IR） of low TS protein group cells for low, middle and high doses to 5-Fu was 0.683 ± 0.131, 0.769 ± 0.114 and 0.836 ± 0.094, respectively. The IR of high TS protein group for low, middle and high doses to 5-Fu was 0.370 ± 0.157, 0.548 ± 0.018 and 0.638 ± 0.020, respectively. IR of 5-Fu tended to be lower in the cells with high TS protein expression than those with low TS protein expression （ P 〈 0.05）. Conclusion TS protein expression levels were significantly correlated to 5-Fu sensitivity in seven pancreatic carcinoma cell lines.
