摘 要:目的探讨癌光啉(PSD-007)在体外对人骨肉瘤MG-63细胞的光动力杀伤效应。方法不同浓度(0、0.5、1.0、2.0、4.0、8.0μg/mL)的PSD-007与细胞孵育4 h后,以不同能量(1.5、3.0、6.0、12.0 J/cm2)激光照射,采用溴化四氮唑蓝(MTT)比色法测定细胞的光密度(D)值及存活率;不同浓度(1.0、2.0、4.0μg/mL)的PSD-007与细胞孵育4 h后,以6.0 J/cm2能量的可见光照射,于光学显微镜下观察光动力治疗后细胞形态的变化,应用膜联蛋白V(Annexin V)/碘化丙啶(PI)双染色流式细胞仪分析细胞死亡形式。结果当PSD-007浓度为2.0μg/mL、光照能量为6.0 J/cm2时,细胞在光动力治疗处理后的存活率明显降低,为52.94%。光动力治疗处理后的细胞逐渐变圆,体积变小,核质比增大,折光性减弱,贴壁能力下降,细胞间隙增大,直到细胞漂浮死亡。流式细胞仪分析结果显示,光动力治疗后死亡细胞主要为坏死或晚期的凋亡细胞。结论在体外,PSD-007对人骨肉瘤MG-63细胞具有光动力杀伤效应。Objective To investigate the photochemotherapeutic effect of PSD-007 on human osteosarcoma MG-63 cells in vitro.Methods MG-63 cells were treated with different concentrations of PSD-007(0,0.5,1.0,2.0,4.0,and 8.0 μg/mL) for 4 hours and were irradiated with different intensites of laser(1.5,3.0,6.0,12.0 J/cm2).Then the D values and survival rates of MG-63 cells were measured by MTT assay.After treatment with different doses of PSD-007(1.0,2.0,and 4.0 μg/mL) for 4 hours and irradiation with 6.0 J/cm2 laser,the morphological changes of MG-63 cells were observed under microscope,and the cell death was determined by Annexin V/PI double staining technique and analyzed by flow cytometry.Results The survival rate of MG-63 cells decreased to 52.94% after PSD-007 treatment at the concentration of 2.0 μg/mL and a light energy of 6.0 J/cm2.After PSD-007 treatment,the cells gradually became round,shrunk,with increased cytoplasm and decreased refractivity and adhesion ability till they died;flow cytometry analysis showed that the cell death was mainly due to necrosis cells and late stage of apoptosis.Conclusion PSD-007 has a photodynamic effect on human osteosarcoma cell line MG-63 in vitro.(Shanghai Med J,2010,33: 558-561)
