摘 要:目的观察光敏剂癌光啉(PSD-007)对小鼠骨肉瘤LM-8细胞的体外光动力效应。方法将不同浓度的光敏剂PSD-007(0.5、1.0、2.0、4.0、8.0μg/mL)与小鼠骨肉瘤细胞株LM-8共同孵育4 h,再以630 nm波长的激光为光源,采用不同激光能量照射LM-8细胞(激光波长为630 nm,光照能量分别为1.5、3.0、6.0、9.0 J/cm2),与空白对照组(不加光敏剂且不接受激光照射)、暗毒性组(加光敏剂但不接受激光照射)、激光组(不加光敏剂但接受激光照射)比较,24 h后采用溴化四氮唑蓝(MTT)法测定其光密度值(D540)值,计算抑制率,并于光学显微镜下观察光动力疗法(PDT)后24 h细胞形态的变化。结果单独照光或光敏剂孵育均无杀伤效应,光敏剂浓度及激光能量是影响杀伤效应的重要因素。PSD-007浓度为4.0μg/mL、光照能量为6.0 J/cm2时,对LM-8细胞有明显的光动力杀伤效果,抑制率为73.8%。光学显微镜下可见细胞形态发生明显的变化,呈坏死或凋亡样改变。结论在体外,PSD-007对小鼠骨肉瘤LM-8细胞具有明显的光动力杀伤作用。PDT是很有前景的治疗骨肉瘤的新方法。Objective To observe the photodynamic effect of PSD-007 on mouse osteosarcoma cell line LM-8 cell in vitro.Methods Mouse osteosarcoma cell line LM-8 was incubated with a serial concentration of PSD-007(0.5,1.0,2.0,4.0,and 8.0 μg/mL)for 4 h,then the cells were irradiated with 630 nm laser at various doses(1.5,3.0,6.0,and 9.0 J/cm2).Another 3 groups(photosensitizer without irradiation,irradiation without photosensitizer,without both) served as controls.Cell inhibition rate was measured by MTT assay 24 h after photodynamic therapy(PDT).Cell morphological changes were investigated under microscope.Results Laser irradiation alone or PSD-007 treatment alone showed no antitumor effect against LM-8 cells.The cytotoxic effects of PDT on the cells were dependent on the PSD-007 concentration and the dose of laser irradiation.PSD-007 at 4.0 μg/mL and laser at 6.0 J/cm2 induced a strong antitumor effect,with the inhibition rate of cells being 73.8%.The cell morphology underwent obvious changes under microscope,showing the morphology of necrosis and apoptosis.Conclusion PDT combined with PSD-007 shows a prominent cytotoxic effect against mouse LM-8 cells in vitro.PSD-007-mediated PDT might be a promising therapy for osteosarcoma.(Shanghai Med J,2010,33: 562-565)
